Rhoifolin Ameliorates Osteoarthritis via Regulating Autophagy

Abstract: Osteoarthritis (OA) is a common age-related joint disease. Its development has been generally thought to be associated with inflammation and autophagy. Rhoifolin (ROF), a flavanone extracted from Rhus succedanea, has exhibited prominent anti-oxidative and anti-inflammatory properties in several diseases. However the exact role of ROF in OA remains unclear. Here, we investigated the therapeutic effects as well as the underlying mechanism of ROF on rat OA. Our results indicated that ROF could significantly allev… Show more

“…Similarly, Rong et al used LPS (0.5 μg/ml) to induce the RAW264.7 cell inflammation model, and detected the autophagy induced by LPS at different times (0–48 h) by Western blot, and found that the expression of LC3/p62 protein was reduced after 24 h of induction ( Rong et al, 2021 ). It has been reported that compared with normal chondrocytes, rat chondrocytes treated with 10 ng/ml IL-1β for 24 h have observed a reduction in autophagy levels ( Yan et al, 2021 ). Interestingly, we found that IL-1β (10 ng/ml) induced ATDC5 chondrocytes for 24 h, and the autophagy level was significantly reduced, which is consistent with previous studies ( Zhou et al, 2021 ).…”

“…Yan et al showed that ROF significantly blocked phosphorylation of P38/JNK and PI3K/AKT/mTOR pathways to enhance autophagy and protected rat chondrocytes from IL-1β-induced inflammation and apoptosis. In vivo , intra-articular injection of ROF also significantly improved cartilage damage in a rat OA model, affirming the potential role of ROF in OA treatment by regulating autophagy ( Yan et al, 2021 ). However, the optimal dose of ROF for in vivo application is unclear and needs to be further investigated.…”

“…3-MA is a commonly used autophagy inhibitor ( 18 , 19 ). As shown in Figures 7A, B , the increased autophagy positive regulatory proteins (such as Atg3, Atg7, Beclin-1, and LC3 I/II) and decreased autophagy negative regulatory protein (P62) by OB were reversed after the employment of 3-MA, indicating that the enhanced autophagy process by OB was suppressed after the 3-MA treatment.…”
