Rho Kinase Regulates the Intracellular Micromechanical Response of Adherent Cells to Rho Activation

Abstract: Local sol-gel transitions of the cytoskeleton modulate cell shape changes, which are required for essential cellular functions, including motility and adhesion. In vitro studies using purified cytoskeletal proteins have suggested molecular mechanisms of regulation of cytoskeleton mechanics; however, the mechanical behavior of living cells and the signaling pathways by which it is regulated remains largely unknown. To address this issue, we used a nanoscale sensing method, intracellular microrheology, to examin… Show more

“…Earlier studies showed that oxLDL triggers activation of Rho-GTPases, the key modulators of cytoskeletal structure, formation of the actin network, and cellular biomechanics (10). It was also shown that Rho activation induces cell stiffening (11)(12)(13). We address the hypothesis, therefore, that activation of RhoA and its downstream signaling pathway plays a central role in oxLDL-induced endothelial stiffening.…”

“…After washing, the cells were incubated with tetramethylRhodamine-conjugated Neutravidin kindly provided by P. C. Searson (Johns Hopkins) at a final concentration of 0.025 mg/ml in 1% BSA in PBS. The cells were visualized with a 60ϫ, oil-immersion objective mounted on an inverted epifluorescence microscope (Nikon, Melville, NY) (Kole et al, 2004).…”

“…The analysis of the random movements of microinjected fluorescent nanoparticles has been used previously to monitor changes in the micromechanical properties of the cytoplasm in cells subjected to biochemical stimuli (Kole et al, 2004a). Although effective, nanoparticle microinjection is inefficient and tedious.…”
