Rho-Kinase Inhibition Reduces Myofibroblast Differentiation and Proliferation of Scleral Fibroblasts Induced by Transforming Growth Factor β and Experimental Glaucoma

Abstract: PurposeWe evaluated prevention of transforming growth factor β (TGFβ)–induced transdifferentiation of cultured scleral fibroblasts to myofibroblasts by rho-associated protein kinase (ROCK) inhibitors. Additionally, we tested whether local delivery of ROCK inhibitors reduced scleral fibroblast proliferation in response to chronic intraocular pressure (IOP) elevation.MethodsPrimary human peripapillary sclera (PPS) fibroblasts were cultured and treated with TGFβ to induce myofibroblast transdifferentiation, as de… Show more

“…Gel contraction alleviated by AR12286 was dose-related. These results are similar to findings from a study by Pitha et al [ 35 ]. Three types of ROCK inhibitors, namely, H1152, Y-27632, and fasudil, were applied in scleral fibroblasts.…”

“…Compared with treatment using other ROCK inhibitors, AR12286 treatment requires lower concentrations (250 and 500 nM). A study reported that 5 to 10 μM ROCK inhibitors were required for myofibroblast differentiation [ 35 , 41 ]. However, drug concentrations may prove to be a limitation of AR12286.…”

“…Thus, activation of fibroblasts by TGF-is an important and essential step by which fibrosis develops. 26 Previous studies have shown that the incubation of human lung fibroblasts with the Ang-(1-7) agonist, A779, reduced levels of TGF-and collagen type I. 27 Therefore, a direct effect of Ang-(1-7) on myofibroblast could have accounted for the antifibrotic effects observed.…”

“…Collagen is produced by interstitials fibroblasts that differentiate in myofibroblasts activated by its binding to TGF‐β. Thus, activation of fibroblasts by TGF‐β is an important and essential step by which fibrosis develops . Previous studies have shown that the incubation of human lung fibroblasts with the Ang‐(1‐7) agonist, A779, reduced levels of TGF‐β and collagen type I .…”

“…The methods of ROCKi delivery may affect the site and mechanism of action. Repeated topical applications can achieve delivery of ROCKi drugs to both anterior and posterior segments, however, intravitreal, subconjunctival, and suprachoroidal injection methods may be more effective for delivery to posterior segment components, such as RGCs and optic nerve axons (Isobe et al, 2016;Pitha et al, 2018). Thus, we used subconjunctival injection to target TM cells, RGCs, and optic nerve axons.…”
