rhKGF-2 Attenuates Smoke Inhalation Lung Injury of Rats via Activating PI3K/Akt/Nrf2 and Repressing FoxO1-NLRP3 Inflammasome

Abstract: Smoke inhalation injury is an acute pathological change caused by thermal stimulation or toxic substance absorption through respiratory epithelial cells. This study aims to probe the protective effect and mechanism of recombinant human keratinocyte growth factor 2 (rhKGF-2) against smoke inhalation-induced lung injury (SILI) in rats. The SILI was induced in rats using a smoke exposure model, which were then treated with rhKGF-2. The rat blood was collected for blood-gas analysis, and the levels of inflammatory… Show more

“…In this study, activities of NF-κB and NRF2, as well as expressions of downstream inflammatory cytokines and anti-oxidant enzymes were mainly analyzed in whole lung lysates in vivo. It is well-accepted that both inflammatory cytokines and LPS can activate NF-κB, but inhibit NRF2 in AECs [ 58 , 59 ]. Whether the crosstalk between macrophage Spry4 regulated inflammation, oxidative stress and ALI through other cell populations (e.g., AECs or neutrophils) should be measured in further study.…”

“…Likewise, PPI analysis revealed that core target proteins of PTE were involved in the PI3K/AKT pathway, such as PIK3CA and AKT1. More and more studies have shown that PI3K/AKT exerts anti-oxidative stress effects by regulating Nrf2 ( Fu et al, 2021 ) ( Liu et al, 2022 ). Meanwhile, the PI3K/AKT signaling pathway also plays an important role in cell apoptosis and proliferation ( Wang et al, 2018 ).…”

“…TNF- α can induce the activation of inflammatory mediators, damage granulocytes and endothelial cells, recruit inflammatory cells, and further induce tissue damage (Malaviya et al., 2017 ). Cigarette or wood smoke exposure would likely induce lung injury and alveolar cell apoptosis (Chen et al., 2021 ; Fu et al., 2021 ); however, ZnCl 2 smoke-induced lung alveolar cell apoptosis had not been reported. Therefore, the inhibition of inflammation and apoptosis is important for treatment of ZnCl 2 smoke-induced ALI.…”
