Rhizogenesis and somatic embryogenesis in calli from wild olive (Olea europaea var. sylvestris (Miller) Lehr) mature zygotic embryos

Abstract: In calli from distal and proximal cotyledon segments and from radicles of mature wild olive zygotic embryos, rhizogenesis prevailed during the first and somatic embryogenesis during the second 30 day subculture period. Rhizogenesis was enhanced by low IBA (0.5 and 2.5/xM) and 5.0/zM 2iP. By reducing by half the media salt concentrations and by inserting a 21 day dark period rhizogenesis was also enhanced. Somatic embryogenesis was inhibited by both 2iP and IBA at concentrations higher than 5.0/~M, but was not … Show more

“…Interestingly, the formation of roots were observed in both varieties, all types of explant, on all media as well as in the dark and on the light. That confi rms statement of Orinos and Mitrakos (1991) that rhizogenesis is a labile morphogenic reaction in the sense that it is infl uenced by various factors tested. In our experiment during the whole cultivation period rhizogenesis was the strongest on the medium with 2iP.…”

“…In our experiment during the whole cultivation period rhizogenesis was the strongest on the medium with 2iP. In contrast this cytokinin can inhibit root formation in higher concentration (Orinos and Mitrakos, 1991). The frequent and spontaneous rhizogenesis on media with diff erent combinations of growth regulators and in diff erent grass pea genotypes was also mentioned by Sinha et al (1983).…”

“…(e.g. Rugini 1988;Orinos and Mitrakos 1991;Mitrakos et al 1992;Leva et al 1995;Rugini and Caricato 1995;Shibli et al 2001; Benelli et al described an efficient true-to-type micropropagation (by stem cuttings) protocol for O. europaea, which was confirmed by RAPD analysis. The analyses of genetic variability/stability during the somatic embryogenesis process (or other micropropagation processes) may be a helpful tool to predict if any detected genetic instability could be initiated at an early stage of callus formation/proliferation and thus contribute to the understanding of different rates of genetic instability among genotypes.…”

“…In O. europaea, somatic embryogenesis has been induced mostly in juvenile material from cotyledonary tissues (Orinos and Mitrakos 1991;Leva et al 1995;Trabelsi et al 2003), immature zygotic embryos (Rugini 1988;Leva et al 1995) and mature embryo callus (Orinos and Mitrakos 1991;Mitrakos et al 1992). Only Rugini and Caricato (1995) succeeded in inducing SE from leaf petiole tissues of adult plants using a method which they designated 'double regenerating system' and which requires previous rejuvenation by in vitro shoot culture.…”

“…Chalkidikis. Orinos and Mitrakos (1991) observed the highest embryogenic capacity in calli derived from radicle explants of mature zygotic embryos of wild olive (O. europaea var. sylvestris) after 21 days of culture on inductive medium with high concentration of auxin.…”
