Rhinovirus Uses a Phosphatidylinositol 4-Phosphate/Cholesterol Counter-Current for the Formation of Replication Compartments at the ER-Golgi Interface

Abstract: Similar to other positive-strand RNA viruses, rhinovirus, the causative agent of the common cold, replicates on a web of cytoplasmic membranes, orchestrated by host proteins and lipids. The host pathways that facilitate the formation and function of the replication membranes and complexes are poorly understood. We show that rhinovirus replication depends on host factors driving phosphatidylinositol 4-phosphate (PI4P)-cholesterol counter-currents at viral replication membranes. Depending on the virus type, repl… Show more

“…Fatty acid synthase encoded by FASN is required for CVB3 replication (Wilsky et al 2012). OSBP and SACM1L are required for sterol/PI(4)P exchange at the ERGolgi interface, and rhinovirus and enterovirus, which belong to Picornaviridae like CVB3, use these processes for viral replication (Mesmin et al 2013;Roulin et al 2014;Strating et al 2015). Our screening results are in agreement with these previous reports and give us confidence in our approach.…”

“…5A). Cross-resistance of the 3A[H57Y] mutant to PI4KIIIb inhibitors and OSW-1 suggests that PI4KIIIb and OSBP act in the same pathway and is in line with previous reports that OSBP is recruited by PI4KIIIb-generated PI4P (Arita, 2014;Arita et al, 2013;Roulin et al, 2014).…”

“…Replication organelles of enteroviruses are enriched in PI4P lipids produced by PI4KIIIb (Hsu et al, 2010). Recently, oxysterol-binding protein (OSBP), a PI4P-binding protein that shuttles cholesterol between membrane compartments, was implicated as another host factor for enterovirus replication (Arita, 2014;Arita et al, 2013;Roulin et al, 2014;Wang et al, 2014).
