RHC 3024: Antiallergic Activity in vitro and Comparison with Disodium Cromoglycate and Other Antiallergic Agents

Abstract: Abstract. RHC 3024 has been investigated for its antiallergic activity in three in vitro models of anaphylaxis. We have also compared its activity profile in these models with that of disodium cromoglycate (DSCG) and other antiallergic agents. As an inhibitor of antigen-induced release of histamine from rat mast cells RHC 3024 was 4 times more potent than DSCG. In the same model the activity profile of RHC 3024 was identical to that of DSCG in the following respects: loss of inhibitory activity with increasing… Show more

“…In contrast, all the drugs displayed little or no inhibition of A23187-induced histamine release. Our results are in agree ment with those reported by many investiga tors (31)(32)(33)(34)(35). However, there are a few papers demonstrating that DSCG inhibits A23187 induced histamine release from rat mast cells (36,37).…”

“…We have previously reported (14,15] that DSCG (3 uM) routinely used as a positive control inhibited AIR by 45.2 ±5.2% (average ±SE, n = 304) and has an I50 value of 4.5 ±0.5 ¡.iM(average ± SE, n = 96). …”

“…The effects of test compounds on spontaneous (SR) and antigen-induced release of histamine (AIR) from passively sensitized rat mast cells were determined according to the procedure of Khandwala et al [14]. Washed peritoneal cells were passively sensi tized in vitro with rat anti-ovalbumin serum.…”
