RFLP Markers for the Selection of Pollination-constant and Non-astringent (PCNA)-Type Persimmon and Examination of the Inheritance Mode of the Markers

Abstract: We investigated marker-assisted selection of the pollination constant and non-astringent (PCNA) trait and inheritance modes of the marker locus in persimmon (Diospyros kaki Thunb.) using restriction fragment length polymorphism (RFLP) analysis. In two backcross progenies, FU-170 and FU-275, 5 of 71 and 23 of 101 offspring, respectively, were judged as PCNA genotype by the marker phenotype. The astringency type of individual offspring was determined by measuring the size of tannin cells and soluble tannin conte… Show more

“…As in the case of the backcross progeny derived from 'Nishimura-wase', 2 RFLP markers, namely, A1 and A2, were segregated in the FU-275 progeny, which was derived from 'Aizumishirazu', and the accuracy of the selection using the RFLP markers has been confirmed (Kanzaki et al, 2008a). However, unlike the case in 'Nishimura-wase'-derived progeny, the primer pair E4/A2r did not produce any fragments in FU-275 (Fig.…”

“…In the previous report, we demonstrated that the AST locus should be regarded as a polysomic locus; however, it remains to be determined whether the inheritance mode of the locus is tetra-or hexasomic (Kanzaki et al, 2008a). In either case, it is necessary to consider that multiple alleles are located separately on 4 or 6 different homologous chromosomes at the single AST locus.…”

“…These DNA markers could improve selection efficiency in Japanese pear breeding programs. Recently, DNA makers linked to the desirable traits for breeding in other fruit trees also developed (Kanzaki et al, 2008;Nakano et al, 2008).…”
