Revisiting the Photon/Cell Interaction Mechanism in Low-Level Light Therapy

Abstract: Objective: Several reports claim that the enzyme cytochrome c oxidase (CCO) is the primary absorber for redto-near-infrared (R-NIR) light in cells and causal for mitochondrial adenosine triphosphate (ATP) upregulation, and that pulsed R-NIR light has frequent therapeutic effects, which are superior to those of the continuous wave (CW) mode used in low-level light therapy (LLLT). Background data: Convincing evidence that the absorption of R-NIR photons by CCO is involved in mitochondrial ATP upregulations as we… Show more

“…45 Most recently, a number of theories have been proposed that may offer additional insight into the phototransduction processes associated with laser-induced analgesia. 46,47 Pending continued research to further elucidate the best standardized approach, it is apparent that higher range dosimetry is associated with an improved clinical result in PBM-assisted pain management. 48 The result of this review is consistent with PBMT being accepted as an evidenced-based approach for orthodontic pain management.…”

“…This information could justify the fact that mitochondrial ATP synthesis can be stimulated by different wavelengths of LED and laser. 16,17 Based on this premise, and partial results obtained by our research group with patients exhibiting bacterial pneumonia, for which we obtained a reduction in hospital stay from 7 to 2 days, for 9 out of 10 treated patients. 18 We propose to treat patients affected by COVID-19 irradiating their thoracic region daily with R-NIR, with 3 J/cm 2 of radiant exposure.…”

“…1,8,35 Recently, however, that hypothesis has been increasingly challenged, 90,91 and a variety of other potential mechanisms and chromophores have been suggested. 2,[92][93][94][95] This study provides several insights regarding light-mediated release of NO and how it may relate to PBM mechanisms. Our finding that multiple wavelengths of light caused release of intracellularly stored NO provides further support that other mechanisms are in play.…”
