Xanthomonas oryzae pv. oryzae (Xoo) is the most infectious pathogen of rice, which causes bacterial leaf blight (BLB) disease. However, the accumulation of chemical or antibiotic resistance of Xoo necessitate the development of its alternative control. In this study, we biologically synthesize three metal oxide nanoparticles (ZnO, MnO2, and MgO) using rhizophytic bacteria Paenibacillus polymyxa strain Sx3 as reducing agent. The biosynthesis of nanoparticles was confirmed and characterized by using UV-vis spectroscopy, XRD, FTIR, EDS, SEM, and TEM analysis. The UV Vis reflectance of the nanoparticle had peaks at 385, 230, and 230 nm with an average crystallite particle size 62.8, 18.8, and 10.9 nm for ZnO, MnO2, and MgO, respectively. Biogenic ZnO, MnO2, and MgO nanoparticles showed substantial significant inhibition effects against Xoo strain GZ 0006 at a concentration of 16.0 μg/ml, for which the antagonized area was 17, 13, and 13 mm and the biofilm formation was decreased by 74.5, 74.4, and 80.2%, respectively. Moreover, the underlining mechanism of nanoparticles was inferred to be in relation to the reactive oxygen species based on their antibacterial efficiency and the deformity in the cell wall phenomenon. Overall, an attractive and eco-friendly biogenic ZnO, MnO2, and MgO nanoparticles were successfully produced. Altogether, the results suggest that the nanoparticles had an excellent antibacterial efficacy against BLB disease in rice plants, together with the increase in growth parameter and rice biomass. In conclusion, the synthesized nanoparticles could serve as an alternative safe measure in combatting the antibiotic-resistant of Xoo.

Prediction and Characterization of Cationic Arginine-Rich Plant Antimicrobial Peptide SM-985 From Teosinte (Zea mays ssp. mexicana)

Antimicrobial peptides (AMPs) are effective against different plant pathogens and newly considered as part of plant defense systems. From prokaryotes to eukaryotes, AMPs can exist in all forms of life. SM-985 is a cationic AMP (CAMP) isolated from the cDNA library of Mexican teosinte (Zea mays ssp. mexicana). A computational prediction server running with different algorithms was used to screen the teosinte cDNA library for AMPs, and the SM-985 peptide was predicted as an AMP with high probability prediction values. SM-985 is an arginine-rich peptide and composed of 21 amino acids (MW: 2671.06 Da). The physicochemical properties of SM-985 are very promising as an AMP, including the net charge (+8), hydrophobicity ratio of 23%, Boman index of 5.19 kcal/mol, and isoelectric point of 12.95. The SM-985 peptide has amphipathic α-helix conformations. The antimicrobial activity of SM-985 was confirmed against six bacterial plant pathogens, and the MIC of SM-985 against Gram-positive indicators was 8 µM, while the MIC of SM-985 against Gram-negative indicators was 4 µM. The SM-985 interacting with the bacterial membrane and this interaction were examined by treatment of the bacterial indicators with FITC-SM-985 peptide, which showed a high binding affinity of SM-985 to the bacterial membrane (whether Gram-positive or Gram-negative). Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) images of the treated bacteria with SM-985 demonstrated cell membrane damage and cell lysis. In vivo antimicrobial activity was examined, and SM-985 prevented leaf spot disease infection caused by Pst DC3000 on Solanum lycopersicum. Moreover, SM-985 showed sensitivity to calcium chloride salt, which is a common feature of CAMPs.

In this study, the biosynthesized silver nanoparticle composites (AgNPCs) obtained from the polysaccharide extract of the red alga Calliblepharis fimbriata were used to control the infection of Xanthomonas oryzae pv. oryzae bacteria causing blight disease in rice. The effect of AgNPCs was evaluated based on seed germination, growth parameters, and photosynthetic pigments of the Oryzae sativa. The formation of AgNPCs was monitored by UV–visible spectrophotometer and characterized by using XRD, SEM, EDAX, and TEM analysis. The synthesized AgNPCs appear spherical in shape, and the size was measured in the range of 25–30 nm. Further, the aqueous extract of seaweed derived AgNPCs was characterized using FTIR, 1D and 2D 1H NMR, and GC-MS analysis. Among the different treatments, a AgNPCs concentration of 50 μg/mL was found to show a stimulatory effect in seed germination and its related growth parameters over control and untreated seeds. In this study, AgNPCs controlled the bacterial colonization and also enhanced the biochemical characteristics of infected rice plants. Later, EDAX analysis showed that infected and uninfected rice seedlings treated with 50 μg/mL of AgNPCs were free of Ag particles but increased amount of nutrients like P, K, C, S, Mg, and Cl in plants. However, reduction in the Ca and Fe ion concentrations was not below the level of untreated plants. This study concluded that AgNPCs of red alga C. fimbriata have growth stimulation as well as pesticidal properties to control bacterial blight in rice without compromising host growth, a potential substrate for nanopesticide production.
