Pseudomonas aeruginosa is an opportunistic pathogen that orchestrate its disease manifestation via a complex communication system known as quorum sensing (QS). Under the regulation of this complex network of QS system, is a myriad of virulence factors including the flagella and pili that contribute to its motility. To establish an infection in a host, P. aeruginosa must first attach itself to host cells with the assistance of flagella and pili and it would be a mere impossible for P. aeruginosa to initiate and establish an infection in the host without those features. In this study, we investigated the motility of P. aeruginosa under the influence of Piper betle leaves extract by growing the bacteria on a semi solid medium and compared the growth spread of the pathogen against the untreated sample. There are three types of motilities that were being investigated, which were swimming, swarming and twitching. In all three assays, a fraction of ethyl acetate extract of P. betle leaves were found to reduce the motility of P. aeruginosa with greatest defect in swimming at more than 80% reduction as compared to the untreated sample. Meanwhile, the fraction partially decreased the swarming and twitching of P. aeruginosa by 58% and 50% reduction respectively, in comparison to the untreated sample. The exhibited defect in the motility of P. aeruginosa proves the anti-QS property of the fraction of ethyl acetate extract of P. betle leaves and could be developed as a potential therapeutic agent against the pathogen.

Phospholipids (PLs) are important biomolecules that not only constitute structural building blocks and scaffolds of cell and organelle membranes but also play a vital role in cell biochemistry and physiology. Moreover, dietary exogenous PLs are characterised by high nutritional value and other beneficial health effects, which are confirmed by numerous epidemiological studies. For this reason, PLs are of high interest in lipidomics that targets both the analysis of membrane lipid distribution as well as correlates composition of lipids with their effects on functioning of cells, tissues and organs. Lipidomic assessments follow-up the changes occurring in living organisms, such as free radical attack and oxidative modifications of the polyunsaturated fatty acids (PUFAs) build in PL structures. Oxidised PLs (oxPLs) can be generated exogenously and supplied to organisms with processed food or formed endogenously as a result of oxidative stress. Cellular and tissue oxPLs can be a biomarker predictive of the development of numerous diseases such as atherosclerosis or neuroinflammation. Therefore, suitable high-throughput analytical techniques, which enable comprehensive analysis of PL molecules in terms of the structure of hydrophilic group, fatty acid (FA) composition and oxidative modifications of FAs, have been currently developed. This review addresses all aspects of PL analysis, including lipid isolation, chromatographic separation of PL classes and species, as well as their detection. The bioinformatic tools that enable handling of a large amount of data generated during lipidomic analysis are also discussed. In addition, imaging techniques such as confocal microscopy and mass spectrometry imaging for analysis of cellular lipid maps, including membrane PLs, are presented.

Search for an efficient anti-cancer compound of natural origin with well-defined mechanisms of action is an important scientific pursuit today, due to cancer being the second leading cause for the death of affected people. The members of the genus Penicillium are one of the important sources of bioactive compounds. In the present study, Penicillium rubens, isolated from a garden soil in Madurai district of Tamil Nadu, was found to produce a highly promising anti-cancer metabolite. The percentage viabilities of HepG2, HeLa and MCF-7 cancer cells treated with the bioactive fraction (P5) isolated from P. rubens, ranged between 40-50% after 96 h. Apoptosis induction was found to be the major reason for the observed reduction in cancer cell proliferation and cell count which was confirmed by caspase activity, DNA fragmentation, clonogenic assay, cell cycle analysis and LDH assays. The upregulation of proapoptotic Bax, coupled with the downregulation of anti-apoptotic Bcl-2 expressions were confirmed by RT-qPCR and flow cytometry methods. The current study also indicated an upregulation of p53 which further strengthened the apoptogenic property of P5 fraction. Non-toxicity of P5 was demonstrated on normal peripheral lymphocytes. The analysis of P5 fraction through GC-MS indicated the presence of indole-2, 3-(4,4-dimethyl-3-thiosemicarbazone) as one of the major compounds. ARTICLE HISTORY
