Review of Osteosarcoma and Current Management

Abstract: Osteosarcoma is the most common primary malignancy of bone in children and young adults. This tumor has a very heterogeneous genetic profile and lacks any consistent unifying event that leads to the pathogenesis of osteosarcoma. In this review, some of the important genetic events involved in osteosarcoma will be highlighted. Additionally, the clinical diagnosis of osteosarcoma will be discussed, as well as contemporary chemotherapeutic and surgical management of this tumor. Finally, the review will discuss so… Show more

“…A combinational application of neoadjuvant chemotherapy and consequent surgical resection significantly increased the five-year survival rate of patients without metastasis to approximately 60%-70% [3]. However, the outcome is disappointing in metastatic patients, with a five-year survival rate of only 5%-20% [4, 5]. Additionally, severe side effects and drug resistance are common.…”

“…Osteosarcoma is the eighth most common primary malignant skeletal tumor affecting children and adolescent. 1,2 Despite the emergence of neoadjuvant chemotherapy, the 5-year survival for osteosarcoma patients with metastasis or recurrence remains poor. 3 The poor prognosis is due to, at least in part, aggressive biologic behavior and delayed diagnosis.…”

“…Currently the 5‐year overall survival rate is approximately 70% for patients diagnosed with primary non‐metastatic extremity OSA and ESA . However, the prognosis for patients with metastatic OSA or ESA at presentation is poor with a 5‐year event‐free survival (EFS) of less than 30% …”
