Reversible Self-Assembly of Glutathione-Coated Gold Nanoparticle Clusters via pH-Tunable Interactions

Abstract: Nanoparticle (NP) clusters with diameters ranging from 20 to 100 nm are reversibly assembled from 5 nm gold (Au) primary particles coated with glutathione (GSH) in aqueous solution as a function of pH in the range of 5.4 to 3.8. As the pH is lowered, the GSH surface ligands become partially zwitterionic and form interparticle hydrogen bonds that drive the self-limited assembly of metastable clusters in <1 min. Whereas clusters up to 20 nm in size are stable against cluster-cluster aggregation for up to 1 day, … Show more

“…Lowering the pH allows GSH to get converted to its zwitterionic form that initiates self-aggregation. [10][11][12] All these factors are sensible for the functioning of the well-segregated GSH-AuNCs in the biological medium. Only under such conditions it becomes feasible to exploit their photocatalytic and photosensitizing properties in photodynamic therapy (PDT).…”

“…Controlled drug release can be achieved by: i) passive targeting through accumulation based on enhanced permeability and retention effect (EPR) observed for relatively large sized nanoparticles (⁓15-100 nm) [13][14][15][16][17] in solid tumor models or ii) active targeting using various payload release mechanism including pH change [18,19] or triggering by endogenous glutathione utilizing thiol-tothiol ligand exchange [20][21][22]. Because ligand exchange is based upon competitive affinity to gold surface between original and incoming ligands [21] MTX can be used not only as a drug but also as a potential targeting ligand [10] because can be directly bound to AuNPs via carboxylic group to form conjugates MTX-AuNPs.…”

“…There are also some new methods for measuring pH, such as colorimetry, fluorescence, electrochemistry, etc. For example, Wang's group designed efficient dual‐mode colorimetric/fluorescence sensor based on carbon dots (CDs) for detection of pH .…”
