This review focuses on recent advances in the development of functionalizable antifouling coatings and their applications in label-free optical biosensors. Approaches to the development of antifouling coatings, ranging from self-assembled monolayers and PEG derivatives to ultra-low-fouling polymer brushes, are reviewed. Methods of preparation and characterization of antifouling coatings and the functionalization of antifouling coatings with bioreceptors are reviewed, and the effect of functionalization on the fouling properties of biofunctional coating is discussed. Special attention is given to biofunctional coatings for label-free bioanalysis of blood plasma and serum for medical diagnostics.

The ultimate goal in protein de novo design is the creation of novel macromolecules with tailor‐made receptor, sensory, and catalytic functions. Despite considerable progress in understanding basic rules of secondary structure formation and protein stability, the well‐known protein folding problem is still far from being solved and, in general, only a limited number of designed proteins are folded uniquely. In this article the state‐of‐the‐art in protein design is demonstrated on some selected examples, indicating that the construction of protein‐like macromolecules mimicking some essential features of natural proteins seems to be within reach. Thus, protein design and mimicry has become an interdisciplinary challenge with most intriguing perspectives. © 1998 John Wiley & Sons, Inc. Biopoly 47: 63–73, 1998

An efficient method for labeling individual proteins in live cells is required for investigations into biological mechanisms and cellular processes. Here we describe the preparation of small quantum dots (QDs) that target membrane surface proteins bearing a hexahistidine-tag (His6-tag) via specific binding to an nitrilotriacetic acid complex of nickel(II) (NTA·Ni 2+ ) on the QD surfaces. We showed that the NTA·Ni 2+ -QDs bound to His-tag functionalized beads as a cellular mimic with high specificity and that QDs successfully targeted His6-tagged vesicle-associated membrane proteins (VMAP) on cell surfaces. This strategy provides an efficient approach to monitoring synaptic protein dynamics in spatially restricted and confined biological environments.
