Reversal of the Hypomethylation Status of Urokinase (uPA) Promoter Blocks Breast Cancer Growth and Metastasis

Abstract: Metastasis is a leading cause of mortality and morbidity in cancer. Urokinase (uPA), only expressed by the highly invasive cancer cells, has been implicated in invasion, metastases, and angiogenesis of several malignancies including breast cancer. Because uPA expression is strongly correlated with its hypomethylated state, we utilized the uPA gene in the highly invasive MDA-231 human breast cancer cells as a model system to test the hypothesis that pharmacological reversal of the uPA promoter hypomethylation w… Show more

“…Accumulating data from the last several years have led to the hypothesis that hypomethylation plays a role in activating certain genes required for metastasis and invasion of a few cancer types other than endometrial cancer. 15,24,47 Results from our study in endometrial cancer add further evidence in favor of the idea that aberrant DNA hypomethylation contributes to the phenotype of more poorly differentiated carcinomas. Methylation-specific PCR analysis of S100A4 gene methylation.…”

“…We examined the effects of CSE on the promoter methylation status of RASSF1A and O 6 -methylguanine-DNA methyltransferase (MGMT) as well as a prometastatic oncogene urokinase-type plasminogen (uPA) (Pakneshan et al, 2004), along with SNCG in A549 cells using the method of multiplex, nested, two-step MSP (MNMSP). The MSP products obtained from step 2 of the nested MSP for the four genes are shown in Figure 3.…”
