Reversal of pancreatic desmoplasia by a tumour stroma-targeted nitric oxide nanogel overcomes TRAIL resistance in pancreatic tumours

Abstract: ObjectiveStromal barriers, such as the abundant desmoplastic stroma that is characteristic of pancreatic ductal adenocarcinoma (PDAC), can block the delivery and decrease the tumour-penetrating ability of therapeutics such as tumour necrosis factor-related apoptosis-inducing ligand (TRAIL), which can selectively induce cancer cell apoptosis. This study aimed to develop a TRAIL-based nanotherapy that not only eliminated the extracellular matrix barrier to increase TRAIL delivery into tumours but also blocked an… Show more

“…Hydrogel is an extremely hydrophilic polymer material with a three-dimensional network structure [ 1 ]. As an excellent water absorption and water retention material, hydrogel has attracted considerable attention in the past few years and has been widely used in the fields of food [ 2 , 3 ], cosmetics [ 4 ], tissue engineering [ 5 ], and drug delivery [ 6 ] as a typical natural polymer.…”

“…The delivery of NO to the PDAC tumor stroma resulted in the reprogramming of activated pancreatic stellate cells, alleviation of tumor desmoplasia, and suppression of antiapoptotic Bcl-2 protein expression, resulting in facilitated tumor penetration by TRAIL and enhanced TRAIL antitumor efficacy in three-dimensional spheroid cultures in vitro and in orthotopic PDAC models in vivo. Thus, the co-delivery of TRAIL and NO by a stroma-targeted nanogel that remodels the fibrotic tumor microenvironment and suppresses tumor growth is promising for PDAC treatment [ 89 ].…”

“…A PEGylated human recombinant TRAIL named TLY012 ameliorated skin fibrosis, pancreatic disease and cirrhosis in preclinical models [ 211 ] and is being actively investigated for anticancer effects in tumor models but is yet to be tested in TNBC models [ 212 ]. Recently, a nanogel delivering nitric oxide and TRAIL to pancreatic ductal adeno carcinoma xenografts had enhanced tumor penetration and efficacy [ 213 ]. A paclitaxel (PTX)-bound albumin nanoparticles with embedded TRAIL (TRAIL/PTX HSA-NP) formulation based on paclitaxel-bound albumin nanoparticles embedded with TRAIL showed improved TRAIL efficacy both in vitro and in vivo pancreatic cancer models [ 214 ].…”
