Reversal of elution order for profen acid enantiomers in packed‐column SFC on Chiralpak AD

Abstract: Enantiomeric separations of four 2-substituted propionic acid drugs have been studied using packed-column supercritical fluid chromatography (SFC) with amylose tris(3,5-dimethylphenylcarbamate) coated on silica as support (Chiralpak AD). Under standard conditions (i.e., flow rate, 1.5 ml/min; column temperature, 30 degrees C; back-pressure, 150 bar), the order of elution could be reversed when the polar alcohol modifier methanol in carbon dioxide was replaced by 2-propanol for ibuprofen, ketoprofen, and naprox… Show more

“…case of 7a and 7b, analytical HPLC employed Chiralpak AD-H and semipreparative SFC employed Chiralpak AS-H. Reversal of elution order is neither uncommon nor predictable, [85][86][87][88] and if only UV detection is employed for analysis or enantiomeric preparative separation, without follow-up using measured CD, OR, or VCD, erroneous conclusions can result with respect to correlation of biological activity and (preliminary) assignment of absolute stereochemistry based on HPLC or SFC elution order.…”

“…3,28 Elution order reversal and enantioselectivity is influenced by both modifiers and chiral selectors used in enantioseparations. 29 The following discussion of three examples highlight the importance of elution order in relation to enantioselectivity and accurate % ee values; elution order reversal was observed on multiple columns across several modifier conditions.…”

“…[256][257][258] A note of caution is appropriate as reversal of elution order of enantiomers is neither uncommon nor currently predictive in HPLC and SFC separations. [259][260][261][262] Therefore, spectroscopic characterization of the eluted enantiomers is paramount to successfully correlating configuration with eutomer (biologically active enantiomer) and distomer (biologically inactive enantiomer).…”
