Reversal of 5-fluorouracil resistance by EGCG is mediate by inactivation of TFAP2A/VEGF signaling pathway and down-regulation of MDR-1 and P-gp expression in gastric cancer

Abstract: The effect of 5-fluorouracil (5-FU) chemotherapy for gastric cancer (GC) is limited by drug-resistance. To conquer this drug-resistance, various treatments including combination therapy have been used, but the overall survival has not been improved yet. In our current study, 5-FU resistant GC cells, SGC7901/FU and MGC803/FU, were established by long term exposure to 5-FU, and the proliferation capability of these resistant cells was verified to be reduced. The drug related proteins, MDR1 and P-gp were up-regul… Show more

“…Studies have confirmed that chemotherapy resistance of cancer cells may also be caused by multi-drug resistance gene (MDR-1) and its encoded protein P-gp. 35 Our previous studies have also confirmed that emodin affected the expression of MRP and P-gp expression levels. P-gp, encoded by MDR, is a transmembrane protein and belongs to the ATP-binding cassette transporter protein superfamily.…”

“…Interestingly, this modulation was reversible. Tang and coworkers found that EGCG suppressed the MDR-1 expression in resistant cells (Tang et al, 2017). The study by Zhu and coworkers revealed that tea polyphenol could improve the uptake of 99mTc-tetrofosmin (a substrate for P-gp) in MCF-7/Adr cells (Zhu et al, 2001).…”

“…Human gastric adenocarcinoma cell line (SGC-7901), maintained in the Surgical Laboratory at Guangzhou Medical University, and the MDR variants SGC-7901/5-FU and SGC-7901/DDP were obtained and maintained in laboratory conditions as described previously [16,17]. Wound healing assay Cells were seeded into six-well plates and transfected with the negative control siRNA-NC or siRNA-XLOC_006753 for 36 h. Then, the cells were serum-starved for 22 h and replenished with 10% fetal bovine serum RPMI-1640 medium.…”
