Rev-erbβ Regulates the Expression of Genes Involved in Lipid Absorption in Skeletal Muscle Cells

Abstract: Rev-erb␤ is an orphan nuclear receptor that selectively blocks trans-activation mediated by the retinoic acid-related orphan receptor-␣ (ROR␣). ROR␣ has been implicated in the regulation of high density lipoprotein cholesterol, lipid homeostasis, and inflammation. Reverb␤ and ROR␣ are expressed in similar tissues, including skeletal muscle; however, the pathophysiological function of Rev-erb␤ has remained obscure. We hypothesize from the similar expression patterns, target genes, and overlapping cognate sequen… Show more

“…The top differentially expressed genes ( Table 2 ) included FRZB (frizzled-related protein B), a negative regulator of canonical Wnt signaling, which, based on two probe sets, had 2.5-fold higher expression in diet-sensitive subjects. Expression of NR1D2 (REV-ERB ␤ ), a transcriptional silencer that regulates expression of lipid uptake genes in skeletal muscle ( 17 ), was signifi cantly decreased in skeletal muscle of diet-sensitive subjects, as was expression of ATP1B1 (Na/K-transporting ATPase ␤ -1 chain) and SLC24A3 (sodium/potassium/calcium exchange member 3). GSEA revealed upregulation of gene sets in ODS versus ODR subjects relevant to Mean (SD).…”

“…The Nr1d2 gene, up-regulated in HF, is highly expressed during daylight and represses Bmal1 transcription (Guillaumond et al 2005). It is also implicated in the regulation of many genes involved in lipid homeostasis in skeletal muscle cells (Ramakrishnan et al 2005). The Tef and the Hlf gene, both up-regulated in HF, serve as mediators in transmitting circadian signals and in controlling various metabolic pathways and physiological functions.…”

“…RORa activates and REV-ERBa represses genes encoding apolipoprotein C-III (ApoC-III) and ApoA1, key proteins in plasma triglyceride and lipoprotein metabolism [60]. RORa and REV-ERBb also regulate many genes involved in lipid homeostasis in skeletal muscle cells [62,63]. REV-ERBa was shown to regulate circadian expression of plasminogen activator inhibitor type 1 (PAI-1), suggesting a role in thrombosis [64].…”
