“…As expected, the anti-tumor effect of CIK depends on the number of active CIK cells and on the overall functional quality of the cells, a factor that is particularly important when considering the techniques employed to culture CIK cells for therapeutic delivery. Recent studies have reported that RetroNectin can improve conglutination, extension, differentiation and proliferation of cultured cells, all of which contribute to more efficient stimulation of T cells (21)(22)(23)(24). Our previous study also confirmed this (25).…”

“…For clinical applications, transduction protocols usually include the human fibronectin fragment CH-296 (also called Retronectin). However, the use of this material is cumbersome both practically and for precise dosage of the additive (coating step), and Retronectin-coated surfaces may reduce the yield of cells obtained after transduction (28). Therefore, the identification of safe, soluble, and easy to manipulate additives with a potent capacity to promote target cell transduction with a broad spectrum of lentiviral pseudotypes, including clinical-grade VSV-G-LVs, remains highly necessary.…”

“…Closed-system bag transduction is advantageous because it reduces the risk of contamination and allows for production on a larger scale. Clinical scale GMP-transduction of primary human T lymphocytes using RN had previously been reported [49], in which T cells were transduced under static condition in the presence of vector containing supernatants or RBV-Spin method. Our improved method has the benefit that T cells were transduced in their own growth medium and retroviral vectors adsorbed on both top and bottom surface of the bag were efficiently utilized.…”
