Retrograde Carbon Monoxide Is Required for Induction of Long-Term Potentiation in Rat Superior Cervical Ganglion

Abstract: Carbon monoxide (CO), produced in the body by the enzyme heme oxygenase (HO), has been suggested as a retrograde synaptic messenger with a prominent role in the long-term potentiation (LTP) of certain areas of the brain. LTP of sympathetic ganglia is 5-HT 3 receptor-dependent and has been shown to require nitric oxide for the maintenance, but not for the induction, phase. We investigated the possibility of CO being required for the induction of ganglionic LTP. Pretreatment of rat isolated superior cervical gan… Show more

“…No apparent action of CO on basal hepatic vascular tone in the present study might be attributed possibly to perfusate free Hb released through hemolysis. It is reported that oxyhemoglobin, oxygenated Hb, inhibits the action of CO (1,9,26,29). The Masterflex roller pump used in the present study should have caused inevitable hemolysis, resulting in release of free Hb in the perfusate.…”

“…Liu et al (38) reported that in the hippocampus, nanomolar concentrations of A␤40 and A␤42 block the response of ␣7-containing nicotinic receptors, which are thought to mediate synaptic currents (39,40) and modulate transmitter release (41,42), thereby contributing to spatial memory and working memory formation. In addition, the ␤-APP holoprotein inhibits heme oxygenase-produced carbon monoxide (7), which has been reported to alter long-term potentiation (43)(44)(45). The role of ␤-APP holoprotein is supported also by findings showing increased mRNA for ␤-APP 751 and ␤-APP 770 in the brains of aged rats with spatial memory deficit but not in rats without impairments (46).…”

“…In that it is a gas, carbon monoxide can travel freely through intracellular and extracellular compartments. The CO literature is vast and complex with many controversies that have yet to be resolved [58][59][60][61][62][63] . CO is better known to be toxic at high levels.…”
