Retracted:Lmx1b-Controlled Isthmic Organizer Is Essential for Development of Midbrain Dopaminergic Neurons

Abstract: The LIM homeodomain transcription factor Lmx1b has been suggested to be required for the differentiation of midbrain dopaminergic (mDA) neurons. However, whether the loss of mDA neurons in Lmx1b Ϫ/Ϫ mice is due to its intrinsic role in the mDA lineage or to a consequence of the malformations caused by the earlier mid/hindbrain patterning defects remains to be clarified. We report here that Lmx1b expression in mDA neurons is dispensable for their differentiation and maintenance, and the loss of mDA neurons in L… Show more

“…Parkinson's disease is a neurodegenerative disorder that involves specific degeneration of dopamine neurons within the substantia nigra regions of the brain. During embryogenesis, dopamine neurons of the substantia nigra arise from the developing ventral midbrain, also known as the ventral mesencephalon, and can be identified by the combination of transcription factors some of which are: Pitx3, Lmx1a, Nurr1, En1, Foxa2 [31][32][33][34][35][36] . Dopamine neurons also arise in the developing forebrain regions; however, these neurons show a different transcription expression profile during development [37] .…”

“…2008). This conclusion is also supported by the fact that normal midbrain dopaminergic neurons are present in Lmx1b −/– mice when the inductive activity of isthmic organizer is rescued by Wnt1 ‐driven Lmx1b expression in the isthmus and when Lmx1b in specifically deleted in differentiating dopaminergic neurons (Guo et al . 2008).…”

“…Furthermore, misexpression of Lmx1b induces ectopic Fgf8 expression non-cell-autonomously by inducing Wnt1, but it represses Fgf8 expression in the isthmus cell-autonomously, possibly mediated by Grg4 in chick embryos (Matsunaga et al 2002). In addition, expression of Lmx1b transgene by Wnt1 promoter in Lmx1b -/mice is able to restore Fgf8 and Wnt1 expression as well as normal development of tectum and cerebellum (Guo et al 2008). Thus, Lmx1b is required and sufficient for the initiation of Fgf8 expression in the early formation of isthmic organizer, and then Lmx1b may restrict the Fgf8 expression domain to anterior limit of the hindbrain by repressing Fgf8 expression cell-autonomously and inducing Fgf8 non-cellautonomously.…”
