This study aims to determine whether pterostilbene improves glycaemic control in rats showing insulin resistance induced by an obesogenic diet. Rats were divided into 3 groups: the control group and two groups treated with either 15 mg kg(-1) d(-1) (PT15) or 30 mg kg(-1) d(-1) of pterostilbene (PT30). HOMA-IR was decreased in both pterostilbene-treated groups, but this reduction was greater in the PT15 group (-45% and -22% respectively vs. the control group). The improvement of glycaemic control was not due to a delipidating effect of pterostilbene on skeletal muscle. In contrast, GLUT4 protein expression was increased (+58% and +52% vs. the control group), suggesting an improved glucose uptake. The phosphorylated-Akt/total Akt ratio was significantly enhanced in the PT30 group (+25%), and therefore a more efficient translocation of GLUT4 is likely. Additionally, in this group the amount of cardiotrophin-1 was significantly increased (+65%). These data suggest that the effect of pterostilbene on Akt is mediated by this cytokine. In the liver, glucokinase activity was significantly increased only in the PT15 group (+34%), and no changes were observed in glucose-6-phosphatase activity. The beneficial effect of pterostilbene on glycaemic control was more evident with the lower dose, probably because in the PT15 group both the muscle and the liver were contributing to this effect, but in the PT30 group only the skeletal muscle was responsible. In conclusion, pterostilbene improves glycaemic control in rats showing insulin resistance induced by an obesogenic diet. An increase in hepatic glucokinase activity, as well as in skeletal muscle glucose uptake, seems to be involved in the anti-diabetic effect of this phenolic compound.

BackgroundMelastoma malabathricum (MM) Linn leaves traditionally use in the treatment of diabetic conditions. The aim of the present investigation was to evaluate the antioxidant, antihyperlipidemic and antidiabetic activity of methanolic extract taken from Melastoma malabathricum Linn (Melastomaceae).MethodsThe methanolic leaves extract of MM Linn leaves used for the study. Chemical test of different extract, acute toxicity study and oral glucose test was performed. Diabetes was induced in rat by single intra-peritoneal injection of streptozotocin (55 mg/kg). The rats were divided into following groups: Group I – normal control, Group II (Vehicle) – diabetic control, Group III (STZ-toxic) – MM I (100 mg/kg, p.o.), Group IV – MM II (250 mg/kg, p.o.), Group V – MM III (500 mg/kg, p.o.), Group VI – glibenclamide (10 mg/kg, p.o.). Bodyweight of each rat in the different groups was recorded daily. Biochemical and antioxidant enzyme parameters were determined on day 28. Histology of different organ (heart, liver, kidney, and pancreas) was performed after sacrificing the rats with euthanasia.ResultsThe methanolic extract of MM did not show any acute toxicity up-to the dose of 2000 mg/kg and shown better glucose utilization in oral glucose tolerance test. Orally treatment of different doses of MM leaves extract decreased the level of serum glucose, glycated hemoglobin, glucose-6-phosphatase, fructose-1-6-biphosphate and increased the level of plasma insulin, hexokinase. MM treatment decreased liver malondialdehyde but increased the level of superoxide dismutase, catalase and glutathione peroxidase. In oral glucose tolerance test observed increased utilization of glucose. Streptozotocin induced diabetes groups rat treated with different doses of MM leaves extract and glibenclamide significantly increased the body weight. Histopathology analysis on different organ of STZ (streptozotocin) induced diabetic rat show there regenerative effect on the liver, kidney, heart and pancreas.ConclusionThe antioxidant, antihyperlipidemic and antidiabetic effect of methanolic extract from Melastoma malabathricum Linn suggests a potential therapeutic treatment to antidiabetic conditions.

Empirical evidence suggested that mind-body interventions can be effectively delivered online. This study aimed to examine whether preventive online mindfulness interventions (POMI) for non-clinical populations improve short- and long-term outcomes for perceived-stress (primary) and mindfulness (secondary). Systematic search of four electronic databases, manuscript reference lists, and journal content lists was conducted in 2016, using 21 search-terms. Eight randomized controlled trials (RCTs) evaluating effects of POMI in non-clinical populations with adequately reported perceived-stress and mindfulness measures pre- and post-intervention were included. Random-effects models utilized for all effect-size estimations with meta-regression performed for mean age and %females. Participants were volunteers (adults; predominantly female) from academic, workplace, or community settings. Most interventions utilized simplified Mindfulness-Based Stress Reduction protocols over 2–12 week periods. Post-intervention, significant medium effect found for perceived-stress (g = 0.432), with moderate heterogeneity and significant, but small, effect size for mindfulness (g = 0.275) with low heterogeneity; highest effects were for middle-aged individuals. At follow-up, significant large effect found for perceived-stress (g = 0.699) with low heterogeneity and significant medium effect (g = 0.466) for mindfulness with high heterogeneity. No publication bias was found for perceived-stress; publication bias found for mindfulness outcomes led to underestimation of effects, not overestimation. Number of eligible RCTs was low with inadequate data reporting in some studies. POMI had substantial stress reduction effects and some mindfulness improvement effects. POMI can be a more convenient and cost-effective strategy, compared to traditional face-to-face interventions, especially in the context of busy, hard-to-reach, but digitally-accessible populations.
