Retinoid X Receptor Gamma Control of Affective Behaviors Involves Dopaminergic Signaling in Mice

Abstract: Abnormal signaling by retinoids or n-3 polyunsaturated fatty acids has been implicated in clinical depression. The converging point in activities of these two classes of molecules is transcriptional activation of retinoid X receptors (Rxr). We show here that ablation of Rxrgamma in mice leads to depressive-like behaviors including increased despair and anhedonia, which were accompanied by reduced expression of dopamine D2 receptor in the shell of nucleus accumbens (NAc) and altered serotonin signaling. While a… Show more

“…Importantly, Rxrg -/-mice show increased despair behaviour and another key symptom of depression (anhedonia), which are both reversed by chronic antidepressant treatment. Adenovirus-mediated re-expression of RXR within the NAc also reversed these behaviours, clearly demonstrating a postnatal function for RXR signalling (Krzyzosiak et al, 2010). These findings suggest that altered retinoid signalling could contribute to diseases affecting the nigrostriatal system, such as Parkinson's and Huntington's diseases.…”

“…4). Finally, RA signaling has been implicated in the regulation of affective behaviors and findings from a specific retinoid receptor knockout mouse model suggest that RA signaling is required for fluoxetine's antidepressant actions (Krzyzosiak et al 2010;Bremner et al 2012;Wietrzych-Schindler et al 2011). Neuronal RA signaling is believed to be the result of a homeostatic balance between local RA synthesis and, more importantly, its feedbackregulated, cytochrome P450-(CYP450) mediated degradation (Shimozono et al 2013).…”
