Retinoic acid signaling cascade in differentiating murine epidermal keratinocytes: Alterations in papilloma- and carcinoma-derived cell lines

Abstract: The retinoic acid (RA) signaling pathway was investigated by transient transfection of a chloramphenicol acetyltransferase (CAT) reporter gene construct containing the RA response element (RARE) of the murine (m) RARbeta2 gene into murine primary epidermal keratinocytes (PEK), papilloma-derived SP1 cells, and carcinoma-derived 3P2 cells. Murine PEK transfected in a low-Ca2+ medium (0.05 mM Ca2+) exhibited a strong transactivation of the CATgene after exposure of the cells to 0.1 microM RA. Transactivation of t… Show more

“…This finding is consistent with our results when the cells are cultured in 10% serum. CRABP-II expression is regulated by various factors such as calcium, estrogen, transforming growth factor-h, and other growth factors present in serum (41)(42)(43)(44). Because MycN is a weak transcription factor with only 2-to 3-fold induction of target genes, the effect of MycN on CRABP-II expression may not be obvious in the presence of these growth factors.…”

“…This surprising finding is in contrast to a previous report (14) in which CRABPI was detected in human Caco‐2 cells by Western blotting using the same antibody. However, keratinocytes may be particular since others have also reported difficulties to detect CRABPI using binding studies (36, 39).…”

“…(A)Hairless mice were exposed to 10 J per cm 2 UVA and sacri®ced 3, 8, or 24 h later. Epidermal CRBP-I was extracted and separated in nondenaturing conditions, and identi®ed by referring to the study reported byVettermann et al (1997) andSorg et al (1999), in which retinoidbinding proteins were assayed in the same conditions. (B) Densitometry analysis of blots A performed with a densitometer from Molecular Dynamics and the software ImageQuant.…”
