Retinoic Acid-Induced Golgi Apparatus Disruption in F2000 Fibroblasts: A Model for Enhanced Intracellular Retrograde Transport

Abstract: Retinoic acid (RA) can transform the Golgi apparatus (GA) into a diffuse vacuolar aggregate and increase the toxicity of some immunotoxins that enter into cells by receptor-mediated endocytosis. An ultramorphological study of the RA-induced GA disruption was performed on F2000 fibroblasts. Cultures were treated with 0.11 to 30 µM RA for 7 -180 min. The endocytosis of Limax flavus agglutinin-peroxidase conjugate (LFA), and the interactions between a phorbol ester (PMA) and RA concerning GA disruption, were exam… Show more

“…Although further investigation is required to delineate the mechanism for the RA effect, we suggest that one possibility is that RA binds to RBP (28) and affects interaction with the putative receptor; interestingly, in the context of TTR and RA inhibition of RBP endocytosis, RA binding influences a region on the RBP surface that is also involved in TTR binding (29). An alternative possibility for the RA influence on RBP endocytic transport is RAdependent biophysical membrane changes at the cell surface (30,31); in the context of cellular transport, for example, it has been reported that 9-cis RA can affect the process of phagocytosis (32).…”

“…RA is an anti-metastatic drug that interferes with the function of the Golgi apparatus [32]. Given that most proteins related to cancer metastasis are glycosylated on the Golgi apparatus [33], its destruction should inhibit metastasis.…”
