RETINOBLASTOMA RELATED1 mediates germline entry in Arabidopsis

Abstract: To produce seeds, flowering plants need to specify somatic cells to undergo meiosis. Here, we reveal a regulatory cascade that controls the entry into meiosis starting with a group of redundantly acting cyclin-dependent kinase (CDK) inhibitors of the KIP-RELATED PROTEIN (KRP) class. KRPs function by restricting CDKA;1-dependent inactivation of the Retinoblastoma homolog RBR1. In and triple mutants, designated meiocytes undergo several mitotic divisions, resulting in the formation of supernumerary meiocytes tha… Show more

“…To test if the Rb1‐kd drives overexpression of the germline factors as seen in other organisms, we analyzed the expression levels of Vasa and Nanos 2 , representative germline markers present in various organisms at Day 1 (23–26 hpf). In the sea urchin embryo, Vasa mRNA is maternally present and becomes gradually restricted into the germline after blastula stage, while Nanos2 mRNA expression is more transient and starts after 32‐cell stage in the germline and decreases by Day 2.…”

“…Since both Rb1 transcript and protein appear to be enriched into mesenchymal cells after Day 1 ( Figures 1B and 2A), these results suggest that Rb1 may have specific functions in pigment cells. To test if the Rb1-kd drives overexpression of the germline factors as seen in other organisms, 10,11,26 we analyzed the expression levels of Vasa and Nanos 2, representative germline markers present in various organisms at Day 1 (23-26 hpf). “…DNA methylation is known to be important in modulating recombination rates in the model plant A. thaliana , which is realized in a strong anticorrelation between recombination rates and DNA methylation (Yelina et al , ; Zhao et al , ; Choi et al , ; Dluzewska et al , ; Tock and Henderson, ; Underwood et al , ). In line with that, we found that recombination rate and the level of DNA methylation were significantly anticorrelated, both in M. polymorpha and P. patens .…”

“…Whether the loss of cohesin contributes to the formation of multiple meiocytes is not clear as yet. The germline in plants has to be established post-embryonically and there is accumulating evidence that the specification of meiocytes also involves a substantial reprogramming of chromatin possibly contributing to the repression of mitotic regulators and stem cell genes [50][51][52] . In this context it is interesting to note that the pattern of histone modifications is altered in swi1 mutants 53 .…”
