Retinal biomarkers and pharmacological targets for Hermansky-Pudlak syndrome 7

Abstract: Deletion of dystrobrevin binding protein 1 has been linked to Hermansky-Pudlak syndrome type 7 (HPS-7), a rare disease characterized by oculocutaneous albinism and retinal dysfunction. We studied dysbindin-1 null mutant mice (Dys −/− ) to shed light on retinal neurodevelopment defects in HPS-7. We analyzed the expression of a focused set of miRNAs in retina of wild type (WT), Dys +/− and Dys −/− mice. We also investigated the retinal function of these mice through electroretinography (ERG). We found that miR-1… Show more

“…The paraffin-embedded retina sections were stained with hematoxylin–eosin, and the micrographs were photographed at 40× magnification under a light microscope (Axiovert, Carl Zeiss Inc., Cambridge, UK). IHC analyses were performed on whole retina, as previously described [ 42 ]. Briefly, retinal sections were dewaxed in xylene, hydrated using graded ethanol, incubated in 0.3% H 2 O 2 /methanol solution for 30 min, and then rinsed with phosphate-buffered saline (PBS; Sigma, Milan, Italy).…”

“…The RNA content changes according to the donor cell type [ 33 ], and noticeable qualitative and quantitative differences have been reported between cancer cells and their physiological counterparts [ 34 , 35 , 36 ]. This growing number of experimental observations, including eye cancer [ 12 , 37 , 38 , 39 ] and retinal degeneration [ 40 , 41 , 42 , 43 , 44 ], paved the way for extracellular RNA exploitation for diagnostic and prognostic purposes [ 45 ]. However, the real biological meaning of such RNA signaling among cells in physiological and pathological conditions remains slippery.…”
