Resveratrol relieves gestational diabetes mellitus in mice through activating AMPK

Abstract: BackgroundGestational diabetes mellitus (GDM) is a disease often manifests in mid to late pregnancy with symptoms including hyperglycemia, insulin resistance and fetal mal-development. The C57BL/KsJ-Lepdb/+ (db/+) mouse is a genetic GDM model that closely mimicked human GDM symptoms. Resveratrol (RV) is a naturally existing compound that has been reported to exhibit beneficial effects in treating type-2 diabetes.MethodsIn this study, we investigated the effect of RV on the pregnant db/+ GDM mouse model, and th… Show more

“…These animals typically present with glucose intolerance, insulin resistance, hyperglycemia, increased food intake, and increased weight gain during, but not prior to, pregnancy2223242526273031. However, several recent studies using the model, including our own, have been unable to reproduce this phenotype323337.…”

“…db/+ mice are heterozygous for a point mutation of the leptin receptor, which renders the receptor inactive20. While female mice appear normal in the virgin state, they develop glucose intolerance during pregnancy21222324252627. In addition, offspring of db/+ mice suffer similar consequences to children affected by GDM (obesity, diabetes in later life)2829, and db/+ mothers that are allowed to age after giving birth also show a phenotype that is reminiscent of post-pregnant mothers afflicted by GDM3031.…”

“…Conversely, a meta-analysis of 10 randomized controlled trials did not indicate a significant effect of resveratrol supplementation upon glucose level (41). The mechanism of action of resveratrol in the treatment of diabetes mellitus seems to be multifactorial; resveratrol may have antioxidant properties, increase AMPK activation, and increase internalization of glucose through modulating glucose transporter expression (98)(99)(100).…”
