Resveratrol reduces oxidative stress and cell death and increases mitochondrial antioxidants and XIAP in PC6.3-cells

Abstract: Resveratrol, a polyphenol derived e.g. from red grapes, has been shown to mediate several positive biological actions such as protection of cells against oxidative stress. It can also influence cell signaling, but the mechanisms behind its antioxidant properties are largely unknown. Here we show that RSV reduces oxidative stress and enhances cell survival in PC6.3 cells depending on the concentration. In these cells, RSV increased the levels of antioxidants, SOD2 and TRX2, and of X chromosome-linked inhibitor … Show more

“…b Neuronal PC6.3 cells were treated with different concentrations of RSV and the activity of the PGC-1a promoter measured as above. Values are means ± SD, n = 3. p \ 0.001 for RSV versus C for all concentrations PGC-1a is neuroprotective in dopaminergic neurons 1163 stress [36]. In line with this, we show here that 5-10 lM RSV elevates SOD2 and Trx2 in cultured dopaminergic cells.…”

“…In line with this, we show here that 5-10 lM RSV elevates SOD2 and Trx2 in cultured dopaminergic cells. SOD2 and Trx2 regulate oxidative stress at the level of the mitochondria and these proteins are regulated in brain neurons partly by NF-jB activation that is induced by various signals including XIAP [28,36]. In this study, we observed no change in NF-jB signaling by RSV, suggesting another mechanism of action.…”

“…[22][23][24] SIRT1 has been shown to deacetylate both histones and a wide range of non-histone proteins; thus it is involved in a wide range of functions including endothelial homeostasis, 25,26 apoptosis, 27,28 cell-cycle regulation, transcriptional silencing and oxidative stress. [29][30][31] Several pieces of evidence suggest that SIRT1 acts as a longevity factor in vascular tissue; in particular, its critical role in endothelial homeostasis by Figure 4 The protein level of SIRT1, FOXO3a, P53 in penile tissues. SIRT1 protein decreased in diabetic group and increased in resveratrol-treated group.…”

“…by activating the nuclear factor erythroid-derived 2-related factor 2 (Nrf2) transcription factor pathway. 120,121 Resveratrol as well as quercetin, catechins, and procyanidins have been demonstrated to counteract oxidative stress-induced apoptosis by increasing the level of mitochondrial antioxidant enzymes and modulating specific signalling pathways and kinase activities, [122][123][124][125][126][127][128][129][130][131] which result in a complex regulation of the balance between pro-and anti-apoptotic Bcl-2 family members. [132][133][134][135][136] These activities might be due to the capability of polyphenols to accumulate in mitochondria.…”
