Resveratrol Protects Vascular Endothelial Cells from High Glucose–Induced Apoptosis through Inhibition of NADPH Oxidase Activation–Driven Oxidative Stress

Abstract: HG induces endothelial cell apoptosis through NF-κB/NADPH oxidase/ROS pathway, which was inhibited by resveratrol. Our findings provide new potential therapeutic targets against brain vascular complications of diabetes.

“…Similarly, resveratrol has been shown to protect endothelial cells by inhibiting NADPH-oxidase and attenuating ROS production [112,113]. Resveratrol also counteracts 7-oxo-cholesterol-induced inflammation and NF-κB activation in human macrophages, suggesting an important role of resveratrol to protect against the pro-atherogenic effect of oxysterols [114].…”

“…Other natural compounds (e.g. melatonin and resveratrol) are known to rescue vascular dysfunctions by the inhibition of NADPH oxidase, a major source of ROS, and the activity of antioxidant enzymes such as Cu,Zn SOD, catalase and GSH peroxidase [147,148,149]. …”

“…Liu et al (Liu et al , 2015b) demonstrated that resveratrol protected PC12 cells from HG-induced neurotoxicity via the PI3K/Akt/FoxO3a pathway, and that resveratrol inhibited HG-induced collagen upregulation in cardiac fibroblasts through regulation of TGF-1-Smad3 signaling (Liu et al , 2015a). Chen et al (Chen et al , 2013) declared that resveratrol protected vascular endothelial cells from HG-induced apoptosis through inhibition of NADPH oxidase activation-driven oxidative stress, whereas Mohamed et al (Mohamed et al , 2014) showed that the neuroprotective effect of resveratrol in diabetic cerebral ischemia-reperfused rats was through regulation of inflammatory and apoptotic events. Together, these studies demonstrated that various mechanisms were responsible for the protective effects of resveratrol on oxidative stress injury.…”
