Resveratrol promotes secretion of wound healing related growth factors of mesenchymal stem cells originated from adult and fetal tissues

Abstract: Non-healing wounds have demonstrated aberrant regulation of several growth factors, thus using exogenous growth factors and cytokines in the clinical setting may improve the outcomes of non-healing wounds. Mesenchymal stem cells (MSCs) are the source of growth factors that show beneficial effect in promoting impaired wound healing. Certain culture condition should be developed to stimulate growth factor secretion from stem cell. Resveratrol, a small molecule found to increase MSCs therapeutic effectiveness. Th… Show more

“…The previous study revealed that conditioned media from retinal pigment epithelial (RPE) cells treated with RV has the ability to inhibit human umbilical vein endothelial cells tube formation. RV at 50 µM concentration suppressed VEGF secretion in hypoxic RPE cells through inhibition of CXC-chemokine c b a receptor 4 [5], [6], [9], [10], [11]. This is the first study that investigated the therapeutic effect of the topical application of specifically manufactured stem cell secretome in severe DED model.…”

“…The baby's umbilical cord of length 3 cm was collected after the mother's consent (ethical clearance number: 1293/KEPK/VII/2019). Cells were isolated using the mixed enzymatic-explant method [10]. Cell pellets were suspended in a culture medium composed of α-MEM, 1% penicillin-streptomycin, 1% amphotericin B, 1% non-essential amino acid (NEAA), 5% fetal bovine serum (FBS), and 5 ng/mL basic fibroblast growth factor, then stored in an incubator at 37°C and 5% CO 2 .…”

“…Resveratrol was also shown to exhibit beneficial properties on the human skin—it delays the aging process and reduces hypertrophic scar formation [ 7 , 13 , 14 , 15 ]. As aging and the formation of non-healing wounds are inseparably associated, there is increasing evidence that resveratrol may even promote wound healing [ 16 ].…”

“…We used ADMSCs because they are readily available, easy to grow in vitro , and can secrete a variety of cytokines ( Heo et al, 2019 ; Prakoeswa et al, 2020 ). Recent studies showed that ADMSC-EXOs contain 148 miRNAs ( Zhang et al, 2017 ) and 1,688 proteins, and function in a variety of cells ( Ni et al, 2018 ).…”
