Restriction fragment length polymorphism of hsp70 gene, localized in the RT1 complex, is associated with hypertension in spontaneously hypertensive rats.

Abstract: Previous studies from our laboratory have demonstrated that the intermediate phenotype of thermosensitivity is present in hypertensive mice and rats. Increased expression of hsp70 caused by increased transcription rate was demonstrated in vivo, in organs, and in cultured cells from spontaneously hypertensive rats and hypertensive mice. In this study, a polymorphism of this gene was revealed with BamHl enzyme by using a human hsp70 probe. A 4.4-kb fragment was visualized in normotensive rats (Brown-Norway BN.Ix… Show more

“…A BP quantitative trait locus (QTL) was identified within the differential chromosomal segment of SHR.1N in a set of recombinant inbred strains derived from the same progenitors (SHR and BN.Lx/Cub) as the congenic strain studied here. 29,43,44 In addition, the existence of the QTL was confirmed by generating a congenic strain. 29 A BP QTL within the same region of rat chromosome 20 was also identified in a cross involving New Zealand genetically hypertensive and BN rats.…”

“…Raised BP has direct effects on the vasculature, 24,25 and vessels subjected to greater mechanical and shear stress express heat shock proteins and are more prone to the development of atherosclerosis. 20,22,25,26 In addition, Hsp70 mRNA levels are enhanced in hypertensive animals, 27,28 and there is a greater accumulation of heat stress-induced Hsp70 mRNA in peripheral blood lymphocytes from hypertensive humans compared with that in NT control subjects. 29 Although these findings demonstrate a stress response to hypertension, a definitive causative link between heat shock protein expression/release and the development of vascular disease has yet to be established.…”

“…In the past few years, we have focused on the general stress response of hypertensive patients, animals and cell lines [14][15][16][17][18]. We believe that the abnormal response to various environmental stresses in hypertension is genetically-determined and that genes induced by heat or other stressors, modify the development of hypertension.…”
