Restoration of extensor excitability in the acute spinal cat by the 5-HT2 agonist DOI

Abstract: 1. The decerebrate cat preparation with an intact spinal cord is characterized by a high degree of excitability in extensor motoneuron pools, which is eliminated by acute spinalization. Subtype-specific agonists for serotonin (5-HT) were investigated in terms of their effectiveness in restoring the extensor excitability following spinalization. 2. Our hypothesis was that 5-HT2 receptors have the primary role in enhancement of extensor reflex excitability, whereas 5-HT1A and 5-HT1B/D receptors are relatively un… Show more

“…However, in our studies we measured the calcium PIC (or sodium PIC) within 10 to 15 min after TTX (or Cd 2ϩ ) application, a time that may not be long enough for the longlasting intracellular actions of metabotropic receptors to be reversed. For example, 5-HT 2c receptor activation can have effects that last for 1 h after 5-HT agonist has been removed (Machacek et al 2001; see also Miller et al 1996). In contrast, metabotropic glutamate receptor facilitation of PICs has a shorter lasting action, and is reversed within a few minutes of removal of receptor activation (Delgado-Lezama et al 1999).…”

“…In rats with complete spinal transections, activation of 5-HT 2C receptors increases the ability of animals to perform weight-supported locomotion (Kim et al, 2001). Moreover, the 5-HT 2A/C agonist DOI upregulates of the excitability of extensor motoneurons in acute spinal cats (Miller et al, 1996). Both observations could be explained by the facilitation of plateau potentials in extensor motoneurons.…”

“…Serotonergic (5-HT) neurons of the raphe nucleus and noradrenergic (NE) neurons from the locus coeruleus may contribute much of this basal excitatory input. 19,20 5-HT and NE act on spinal motor neurons yielding plateau potentials, which may amplify excitatory inputs including reflex inputs. [21][22][23] Plateau potentials originate on dendrites and amplify excitatory inputs up to six-fold, thus leading to sustained firing with minimal excitatory input.…”
