Response to Growth Hormone Treatment and Final Height in Noonan Syndrome in a Large Cohort of Patients in the KIGS Database

Abstract: This group of patients with NS showed an early response to GH treatment, with an attenuation of this effect thereafter. At final height the median increment of final height was 0.61 SDS according to Tanner standards and 0.97 SDS according to Noonan standards. No serious side effects were reported.

“…Intriguingly, these doi: 10.7243/2052-8000-2-1 studies may be distinguished in two classes, depending on the extent of the increase of height SDS they report. Thus Kirk et al, observed a moderate increase in height SDS of 0.8 in 10 patients [22], which is in accordance with a rise of 0.87 in 24 patients reported by Raaijmakers et al, [29]. On the other hand, Osio et al, Noordam et al, and Romano et al, reported better outcomes of GH therapy, with increases in height SDS ranging from 1.3 to 1.7 on a total of 112 patients [26,28,30].…”

“…As a whole, current data suggest that the overall increase in height SDS with GH therapy in Noonan syndrome ranges from 0.8 to 1.7 [12,16,30,33], which represents a final height gain of 5 to 10 cm. At first sight, this may seem to be a small gain; however, an increase of 5 to 10 cm may allow a large subset of patients to reach normal adult height.…”

“…The response to GH therapy in NS can be affected by a number of factors, such as dose of rhGH and type of genetic mutation involved. Moreover, results of long-term GH treatment of short stature due to Noonan syndrome demonstrate a significant improvement in adult height [12].…”
