Response of resting human peripheral blood natural killer cells to interleukin 2.

Abstract: The present study shows that recombinant interleukin 2 (IL-2) purified to homogeneity induces a rapid and potent enhancement of spontaneous cytotoxicity of human peripheral blood lymphocytes. The cells mediating cytotoxicity after 18-h treatment with IL-2 have surface markers of natural killer (NK) cells and are generated from the peripheral blood subset containing spontaneous cytotoxic cells. A parallel production of gamma interferon (IFN-gamma) is induced by recombinant IL-2 (rIL-2), and NK cells appear to b… Show more

“…The cytokine network is hence sufficiently established by IFN -production originating from Th1 cells. Nevertheless, NK cells, which predominantly contribute to the early production of IFN -in response to IL -12, 66 are necessary for its full activation. In addition, (3 ) the feedback loop is interrupted in IFN -receptor± deficient mice as evidenced by IL -12 serum levels below 1 ng /mL despite profound IFN -production upon IL-12 DNA injection.…”

“…A major source of IFN-within the population of human leucocytes is NK cells, a subset of lymphocytes with CD3 -/56 + phenotype that constitutively kill tumour cells without apparent restriction by histocompatibility molecules [2]. Resting human NK cells carry the p70 moiety of the IL-2R [3] and respond to IL-2 by induction of IFN-production [4,5] as well as enhanced cytolytic activity and proliferation [5].…”

“…Mature peripheral blood natural killer (NK) cells, similar to lymphocytes, retain some proliferative capacity 11 . NK cells participate in the control of viral or bacterial infection, regulation of hematopoiesis, production of cytokines and cytotoxicity of neoplastic cells.…”
