Response of leptin to short-term and prolonged overfeeding in humans.

Abstract: As one of the postulated roles of the ob gene product, leptin, is regulation of energy balance and preservation of normal body composition, we investigated the effect of acute and chronic calorie excess (weight gain) on serum leptin in humans. Two protocols were employed: 1) acute (12-h) massive (120 Cal/kg) voluntary overfeeding of eight healthy individuals; and 2) chronic overfeeding to attain 10% weight gain, with its subsequent maintenance for additional 2 weeks, involving six normal males. In the acute ex… Show more

“…In adults, circulating leptin predictably rises after overfeeding [36] and falls after fasting [37]. However, whether specific macronutrients alter serum leptin concentrations independent of energy intake is equivocal.…”

“…Several studies have demonstrated that the leptin gene expression is positively regulated by insulin both in vivo (Saladin et al, 1995;Kolaczynski et al, 1996c) and in vitro (Kolaczynski et al, 1996c;Leroy et al, 1996). Plasma leptin levels decrease after fasting and increase by refeeding in humans (Kolaczynski et al, 1996a,b;Boden et al, 1996;Dubuc et al, 1998;Geldszus et al, 1996;Wadden et al, 1998) and rodents (Frederich et al, 1995), which parallels the change in plasma insulin levels (Kolaczynski et al, 1996b). Accordingly, it is possible that acutely decreased insulin levels during the early phase of an LCD may contribute to a rapid fall in plasma leptin levels.…”

“…24 Several pediatric studies have not reported an association between resistin or adiponectin and weight status, leptin or insulin concentrations. 8,25,26 In adults, circulating leptin predictably rises after overfeeding 27 and falls after fasting. 28 However, whether specific macronutrients alter serum leptin concentrations independent of energy intake is equivocal.…”
