Response of isolated rat descending vasa recta to bradykinin

Abstract: Outer medullary descending vasa recta (OMDVR) were dissected from the outer medullary vascular bundles of young rats, perfused in vitro, and loaded with fura 2 for measurement of intracellular calcium concentration ([Ca2+]i) by fluorescent ratio imaging. Fluorescent video images revealed that fura 2 selectively loads into endothelial cells but not pericytes. Bradykinin (BK), at concentrations >10−11 M, elicited an increase in [Ca2+]ifrom baseline values in the range from 50 to 100 nM to peak values of 600–8… Show more

“…As expected, BK generates a peak-and-plateau Ca 2ϩ response, enhances NO generation (Fig. 8), and induces vasodilation (112,129,130). An unexpected finding is that the vasoconstrictor ANG II suppresses basal Ca 2ϩ and inhibits BK-, acetylcholine-, thapsigargin-, and cyclopiazonic acid-induced Ca 2ϩ responses in DVR endothelia (Fig.…”

“…Exposure of HepG2 cells to ketamine for 6 and 24 h significantly decreased the bradykininenhanced calcium mobilization. Bradykinin increases calcium influx through stimulation of the bradykinin-B2 receptor (Pallone et al, 1998). This endogenous bradykinin-B2 receptor is found in hepatocytes (Yayama et al, 2007).…”

“…The focus of these studies has been on the larger diameter vasa recta vessels of the outer medulla for reasons related to the unique structure and function of the medullary vasa recta circulation. Before the results of these studies are reviewed, it is important to first appreciate the microvascular anatomy of the renal medulla, as described in detail by others (3,4,40,44,64,(81)(82)(83)127). In brief, it is recognized that blood flow to the medulla of mammalian kidneys is supplied by postglomerular microvessels derived from juxtamedullary glomeruli, the deep glomeruli of the inner cortex that receive their blood supply from the interlobular branches of the arcuate arteries, as described by Kaissling and Kriz (39).…”

“…For example, these vessels have been shown to respond to vasoconstrictors such as ANG II, as shown in Fig. 3A (85), AVP (124), and endothelin (112), as well as vasodilators such as bradykinin (83), prostaglandin E 2 (112), and adenosine (111). The ability of the DVR to behave like small arterioles accounts for the ability of the medullary flow to be controlled independently from the cortical circulation when vasoactive substances are endogenously released or infused.…”
