Response and Recovery in the Plasma Metabolome Tracks the Acute LCMV-Induced Immune Response

Abstract: Lymphocytic choriomeningitis virus (LCMV) infection of mice is non-cytopathic, producing wellcharacterized changes reflecting host immune response. Untargeted metabolomics using mass spectrometry identified endogenous small molecule changes in blood from mice inoculated with LCMV, sampled at days 1, 3, 7 and 14 post infection. These represent well-characterized time points in acute LCMV infection and the immune response. Diverse pathways were altered, including TCA cycle intermediates, γ-glutamyl dipeptides, l… Show more

“…The formulation of the cell danger hypothesis was based on the recognition that similar metabolic pathways were coordinately regulated as an adaptive response to cellular threat regardless of whether the perturbation was caused by a virus,15 a bacterium,16 genetic forms of mitochondrial disease,10 or neurodevelopmental disorders with complex gene–environment pathogenic mechanisms like autism 17. These metabolic pathways traced to mitochondria.…”

“…The poor quality of these two models was perhaps suggestive of the attempts by the treated animals to regain homeostatic balance or to adapt to prolonged CHT ingestion [26]. It appeared that the rats' response to CHT intervention followed a response and recovery pattern [27]. However, the OPLS-DA model for day 38 could again produce an obvious differentiation between the groups, as indicated by the good Q 2 and permutation testing, which suggested repeated ingestion of CHT produced gradual and cumulative changes in rats.…”

“…Many of the metabolomics studies in nutritional immunology have focused on how different infectious agents and signaling molecules alter the metabolic profile of immune cells (Brugger et al, 2006;Diamond et al, 2010;Wikoff et al, 2009). Metabolomics was used in a lymphocytic choriomeningitis virus (LCMV) infection model to elucidate systemic metabolic changes induced during an acute viral infection (Wikoff et al, 2009).…”

“…Metabolomics was used in a lymphocytic choriomeningitis virus (LCMV) infection model to elucidate systemic metabolic changes induced during an acute viral infection (Wikoff et al, 2009). The metabolites identified had similar kinetic profiles, whereby the metabolite concentration declined until day 7, when the peak viral load was observed, and either returned to baseline when the infection was resolved (day 14) or in some cases, remained low.…”
