“…The contour length of S. cerevisiae mtDNA ranges from 21 to 25 um (51,96), corresponding closely to the molecular mass determination of 46 x 106 daltons derived from sedimentation data (15). The mtDNA of several other yeasts has been reported (89,90) to range in contour length from one-quarter to one-half that of S. cerevisiae.…”

“…In addition, conflicting results have led to the lack of agreement concerning the effects of EB on nucleic acids in living organisms (McMurray and van Holde, 1991). One of the best known applications of EB is the irreversible destruction of mitochondrial genomes in some yeasts and fungi (Slonimski et al, 1968;Goldring et al, 1970;Whittaker et al, 1972;O'Connor et al, 1976;Niagro and Mishra, 1989). Although the molecular mechanism underlying this phenomenon remains unclear, it has been established that EB inhbits de novo synthesis of mitochondrial DNA (mtDNA) as well as stimulating a rapid destruction of pre-existing mtDNA molecules, resulting in a 100% conversion of a wild-type culture into a respiratory-deficient one.…”

“…The medium for the screening of respiratory-deficient mutants was prepared according to O'Connor et al (1976). The complete medium (GYP) contained (per litre) 10 g of Bacto peptone, 20 g of glucose, 1 g of KH 2 PO 4 and 1 g of MgSO 4 Á 7H 2 O. Glycerol medium (GlyYP) was made using 20 g of glycerol instead of 20 g of glucose in medium GYP.…”
