Resolving the mystery—How TCR transgenic mouse models shed light on the elusive case of gamma delta T cells

Abstract: Cutting‐edge questions in αβ T cell biology were addressed by investigating a range of different genetically modified mouse models. In comparison, the γδ T cell field lacks behind on the availability of such models. Nevertheless, transgenic mouse models proved useful for the investigation of γδ T cell biology and their stepwise development in the thymus. In general, animal models and especially mouse models give access to a wide range of opportunities of modulating γδ T cells, which is unachievable in human be… Show more

“…One of the more challenging aspects in this context is the exploration of γδ T cell function [143]. While the murine counterparts have been extensively studied in recent decades [144], strategies to study human cells in the mouse context remain experimentally more demanding. The reconstitution of immunocompromised murine hosts with selected cells of human origin, a process often referred to as "humanization", is a widely used approach to study human cancer, human immune cells, but also many other human cell types under physiologic conditions [145].…”

“…γδ T cells have complex biology and tissue specific tropism; therefore, they may not behave in in vitro culture as they would under physiologic conditions. This makes interpreting the results of γδ T cells from in vitro studies challenging [128]. To harness the γδ T cell anti-tumor activity, many researchers have attempted to activate γδ T cells in vivo for the treatment of a broad range of tumors.…”

“…Further, γδT cells differ from conventional αβT cells, due to their restricted T cell receptor (TCR) diversity (22). Interestingly, when looking at the potential combinations of the variable (V), diversity (D) and joining (J) segments, the γδTCR diversity is significantly higher than both the B cell receptor (BCR) and αβTCR with 10 20 potential combinatorial diversities, in comparison to 10 11 and 10 15 , for BCR and αβTCR, respectively (23,24). Remarkably, the TCR repertoire effectively expressed by γδT cells is greatly limited, with some oligoclonal γδT cell subsets dominating in certain organs.…”
