Resolution of the oedema associated with allergic pulmonary inflammation in rats assessed noninvasively by magnetic resonance imaging

Abstract: 1 Magnetic resonance imaging (MRI) was used to study noninvasively the effects of compounds to resolve inflammation induced by ovalbumin (OVA) challenge in the lungs of actively sensitised rats.

“…Both RPL554 and RPL565 when delivered as a dry powder significantly attenuated plasma protein extravasation induced by histamine in the guinea pig. This is in agreement with previous studies demonstrating that the PDE4-selective inhibitors NVP-ABE171 and YM976 inhibited antigen-induced edema in the airways (Aoki et al, 2001;Tigani et al, 2003). Together, these studies demonstrated the effectiveness of this class of drug on edema formation in the airways.…”

“…Unfortunately, this technique has not been shown to be a useful assessment of airway inflammation in asthmatics [15], unless under conditions of a segmental allergen challenge [16,17], most likely because neutrophils, a cell-type with a debatable association to allergic asthma, use more glucose than other cell types when activated [18,19]. Magnetic resonance imaging has also been used to detect inflammatory changes [20][21][22] as well as functional disruption after allergen exposure [23] in experimental models of asthma. Although this modality provides superior soft tissue contrast with no known deposition of radiation, CT is more widely available and is more commonly associated with PET and single photon emission CT (SPECT) radio-imaging technologies.…”

“…The MRI signals after OVA challenge correlated significantly with a variety of inflammatory parameters determined in the BAL fluid recovered from the same animals. It is noteworthy that the fluid signals detected by MRI correlated significantly with the perivascular edema assessed by histology (Tigani et al, 2003a;Blé et al, 2008). Micro-CT also has been used to quantify edema in a rat model of allergic lung inflammation (Jobse et al, 2009).…”

“…When assessing the effects of anti-inflammatory drugs administered before disease induction in these models, a doserelated reduction of the MRI signals has been shown for compounds such as the glucocorticosteroids budesonide (Beckmann et al, 2001a;Tigani et al, 2003a;Blé et al, 2009a) (Fig. 2) and mometasone (Tigani et al, 2003a) and a mitogen-activated protein kinase inhibitor (Tigani et al, 2003a).…”
