Resolution of Der p1-Induced Allergic Airway Inflammation Is Dependent on CD4+CD25+Foxp3+ Regulatory Cells

Abstract: Allergic airway inflammation (AAI) is characterized by airway hyperreactivity, eosinophilia, goblet cell hyperplasia, and elevated serum IgE, however, it is unclear what mediates natural resolution after cessation of allergen exposure. This is important because the outcome of subsequent allergen challenge may depend on the concurrent inflammatory milieu of the lung. Using a murine AAI model, we demonstrate that after exposure to a defined natural protein allergen, Der p1, the response in lungs and draining med… Show more

“…These cells were able to suppress OVA-specific T cell proliferation ex vivo. Other reports have shown that depletion of CD4ϩCD25ϩ cells before acute airway allergen challenges exacerbated lung eosinophilia, and adoptive transfer of these cells can also significantly protect against developing lung inflammation (26,29). Not surprisingly, mice deficient in Foxp3 have increased lung inflammation and smooth muscle mass after repetitive antigen challenge (10).…”

“…Sections were stained with hematoxylin and eosin (H&E) or Periodic acid-Schiff (PAS). Slides were scored for the presence of goblet cells as previously described (55). Average percentages of goblet cells within 10 consecutive small airways (less than half the width of a field at ×200 magnification) were calculated for each slide, under experimentally blinded conditions.…”
