Resistance in Malignant Tumors: Can Resistance Assays Optimize Cytostatic Chemotherapy?

Abstract: The frequent resistance of malignant tumors to chemotherapy documents the fact that numerous patients are uselessly traumatized by highly toxic drugs. Investigations into this resistance have shown that many different mechanisms exist which can abolish the antitumor action of chemotherapeutics. So far, no safe method has been found to counteract this resistance. To date, the only way to avoid harm by an ineffective chemotherapeutic is to refrain from its use when resistance to it is detected. The present surve… Show more

“…Drug resistance is related to numerous changes in the tumor and its microenvironment 4 and can reflect both tumor and normal tissue. 5 An approach to reduce side effects and drug resistance is the usage of prodrugs. Prodrugs are chemically modified, preferably nontoxic, versions of the active anticancer agent.…”

“…Drug toxicity [2] and tumour resistance to chemotherapeutic agents [3,4,5,6,7] are 2 principal reasons for treatment failure in a great majority of patients with metastatic cancer. Thus, the development of successful approaches to treat cancer will require combinations of drugs and/or treatment modalities to optimise the tumour response to therapy whilst modulating the harmful side effects.…”

“…Despite significant advances in the chemotherapeutic treatments of neoplastic diseases, success in the treatment of many solid tumours has been limited [1]. Drug toxicity [2] and resistance development during therapy remain the 2 principal reasons for treatment failure in a great majority of patients with metastatic cancer [3][4][5]. Chemotherapy is the major therapeutic approaches in several human life-threatening diseases including microbial infections, autoimmune diseases and cancers [6] and drug combinations are the key to most cancer treatments [7][8] Thus, the development of successful approaches to treat cancer will require combinations of drugs and/or treatment modalities to optimize the tumour response to therapy whilst modulating the harmful side effects.…”
