Rescue of Hearing, Auditory Hair Cells, and Neurons by CEP-1347/KT7515, an Inhibitor of c-Jun N-Terminal Kinase Activation

Abstract: We have studied the mechanisms of auditory hair cell death after insults in vitro and in vivo. We show DNA fragmentation of hair cell nuclei after ototoxic drug and intense noise trauma. By using phospho-specific c-Jun-N-terminal kinase (JNK) and c-Jun antibodies in immunohistochemistry, we show that the JNK pathway, associated with stress, injury, and apoptosis, is activated in hair cells after trauma. CEP-1347, a derivative of the indolocarbazole K252a, is a small molecule that has been shown to attenuate ne… Show more

“…Further research into the role of GADD45b in hair cells is needed to substantiate this hypothesis, however. It is also possible that NF-kB is involved in regulation of the cellular antioxidant program in the cochlea, as it has been proposed for neuronal cells and other cell types that harbor NF-kB in a constitutive activated state (Schü tze et al 1992;Kaltschmidt et al 1994;Lezoualch'l et al 1998;Jang and Surh 2004).…”

“…Studies show that reactive oxygen species (ROS) and activated c-Jun N-terminal kinase ( JNK) signaling are some of the key mediators of hair cell death (Clerici et al 1996;Huang et al 2000;Pivola et al 2000;Sha and Schacht 1999). However, surprisingly little is known about the factors that signal hair cell survival.…”

“…prevented by the application of JNK inhibitors (Pirvola et al 2000;Bodmer et al 2002b;Ylikoski et al 2002). Other recent evidence suggests that the intracellular signals leading to JNK activation in hair cells include the small G proteins Ras and Rac/cdc42 (Bodmer et al 2002a).…”

“…There are growing numbers of compounds that have been found to protect the cochlea from gentamicin toxicity (Yokota et al 1989;Garetz et al 1994;Xuan and Dong 1995;Song et al 1998;Gao 1999;Sha and Schacht 1999;Suzuki et al 2000;Sinswat et al 2000;Pirvola et al 2000). It is generally assumed that the protective effects of these compounds are due to direct effects upon hair cells.…”
