Requirement for Slit-1 and Robo-2 in Zonal Segregation of Olfactory Sensory Neuron Axons in the Main Olfactory Bulb

Abstract: The formation of precise stereotypic connections in sensory systems is critical for the ability to detect and process signals from the environment. In the olfactory system, olfactory sensory neurons (OSNs) project axons to spatially defined glomeruli within the olfactory bulb (OB). A spatial relationship exists between the location of OSNs within the olfactory epithelium (OE) and their glomerular targets along the dorsoventral axis in the OB. The molecular mechanisms underlying the zonal segregation of OSN axo… Show more

“…Probes were synthesized from cDNA clones encoding Kirrel1, Kirrel2 and Kirrel3 (kind gifts of Dr David Ginty, Johns Hopkins School of Medicine, Baltimore, USA), and G αo and G αi2 (Cloutier et al, 2002). Fresh frozen brains or nasal cavities were cryosectioned at 20 μm, fixed, and processed as previously described (Cho et al, 2007).…”

“…Neuropilin-2 (Nrp2) and Robo-2 are good candidate guidance molecules determining the D -V axis, because they show graded expression in the OE (Norlin et al 2001;Cho et al 2007). Nrp2 is expressed in a ventral-high and dorsallow gradient, whereas Robo-2 expression shows a dorsal-high and ventral-low gradient in the OE and OSN axon termini (Fig.…”

“…Slit was first identified in Drosophila as a molecule secreted by midline cells, and was later shown to repel the extension of axons expressing Robo receptors (Brose et al, 1999;Kidd et al, 1999;Seeger et al, 1993) and to regulate the migration of cells such as neuronal precursors . In the olfactory system, members of the Slit and Robo families are expressed in a specific spatiotemporal pattern and play important roles in the guidance of olfactory sensory axons (Cho et al, 2007;Marillat et al, 2002;Nguyen-Ba-Charvet et al, 2008;Yuan et al, 1999). Whether OECs, which have the same developmental origin as olfactory sensory neurons, are also responsive to Slits is unknown.…”
