Requirement for natural killer T (NKT) cells in the induction of allograft tolerance

Abstract: Together, these findings indicate that V␣14͞V␤8.2 is a unique antigen receptor for NKT cells but not for conventional T cells. Because V␣14 NKT cell development was inhibited largely in mice that lack the MHC class I-like molecule CD1d, positive selection of V␣14-expressing immature NKT cells requires CD1d expression (14,15). Recently, it has been shown that glycolipid, ␣-galactosylceramide (␣-GalCer), and glycosylphosphatidylinositol-anchored proteins can be presented by murine CD1d (13, 16). Subsequently, it… Show more

“…Many studies have suggested that an important physiological function of NKT cells is to control immune responses against infection and some tumors. In transplantation immunity, CD1d-dependent NKT cells are also thought to play a role in the induction of self 19 or allograft [20][21][22] or xenograft tolerance. 23 In a murine GVHD model, bone marrow DN NK1.1 þ T cells were observed to be important for preventing GVHD.…”

“…Early interest in murine invariant CD1d-reactive T cells was sparked by their ability to rapidly produce large amounts of IL-4 and IFN-␥ after stimulation, 9,10,32 suggesting a role in the development of Th2- 64,65 Finally, invariant CD1d-reactive T cells promote tolerance in murine models of transplantation, such as cardiac allografts, 66 although the specific role of these cells in promoting tolerance of liver allografts is unknown. Therefore, under appropriate conditions, CD1d-reactive T cells can have anti-inflammatory and/or immunomodulatory properties.…”

“…2B). Since previous studies (15,23) indicated that the regulatory functions of NK1.1 ϩ T cells are mediated at least in part by IL-4 or IFN-␥, the survival of heart allografts was compared in IL-4 Ϫ/Ϫ , IFN-␥ Ϫ/Ϫ , and wild-type BALB/c hosts given the complete conditioning regimen. Fig.…”
