Repurposing of an old drug: In vitro and in vivo efficacies of buparvaquone against Echinococcus multilocularis

Abstract: The metacestode stage of the fox tapeworm Echinococcus multilocularis causes the lethal disease alveolar echinococcosis. Current chemotherapeutic treatment options are based on benzimidazoles (albendazole and mebendazole), which are insufficient and hence alternative drugs are needed. In this study, we screened the 400 compounds of the Medicines for Malaria Venture (MMV) Pathogen Box against E. multilocularis metacestodes. For the screen, we employed the phosphoglucose isomerase (PGI) assay which assesses drug… Show more

“…The Pathogen Box project is currently active, but several groups have already reported anthelmintic screens using this library [129,135,[161][162][163][164][165]. We have compiled the results from these published screens in Figure 5.…”

“…C. elegans viability screen -automated quantification of viability by differential absorption of the dyes DB-1 and propidium iodide [164]. E. multilocularis metacestode screen -damage was assessed by quantifying the activity of phosphoglucose isomerase released into the culture media [162]. F. hepatica screens -viability of metacercariae was assessed using a scoring system taking into account membrane damage and fluke translucency, and viability of adults was assessed using a scoring system taking into account worm motility, colour and rigidity [165].…”

“…• Anti-theilerial drugs MMV689480 (buparvaquone) and MMV671636 (ELQ-400) have demonstrated a cytotoxic effect [29]. Buparvaquone impaired parasite mitochondria, and with an IC50 of 2.87 μM and 0.02 μM, respectively, against in vitro cultured E. multilocularis metacestodes, buparvaquone can be an effective therapeutic choice [31].…”

“…Dual activity against Giardia and Cryptosporidium were reported for MMV010576, MMV028694 and MMV676501 that showed ≥ 75% inhibitory efficacy 8 . Several such studies have accelerated the identification of small molecules that are now re-profiled, repurposed and reclassified to show broader inhibitory activities on more than one pathogenic agent 912 .…”
