RESUMEN.Mytilus chilensis tiene ciclos reproductivos que varían latitudinalmente. Presenta reducida diferenciación genética y morfológica debido a un gran potencial de dispersión. Se acondicionaron reproductores de bahía Yaldad (Chiloé) y bahía Zenteno (Punta Arenas) a 9 ± 0,5°C y 15 ± 0,5ºC, alimentados con dieta (1:1) de Isochrysis galbana y Chaetoceros neogracile. Se espera dilucidar si el acondicionamiento a diferentes temperaturas produce variaciones en el potencial reproductivo de las poblaciones. El menor desarrollo gonadal se produjo en los reproductores acondicionados a 9ºC, mientras que el mayor se produjo en los reproductores acondicionados a 15ºC provenientes de Chiloé. La fecundidad de los reproductores de Yaldad fue mayor que los de Zenteno. El diámetro de los ovocitos fue mayor en los reproductores de Zenteno y en ambas poblaciones fue mayor a 9ºC. Ni el porcentaje de huevos fecundados ni el porcentaje de eclosión de larvas D mostraron diferencias significativas entre las poblaciones a ninguna de las temperaturas de acondicionamiento. De acuerdo con estos resultados, no se logra establecer diferencias en el potencial reproductivo en las poblaciones y bajo las condiciones de este estudio. Assessment of the reproductive potential of the mussel (Mytilus chilensis) from two natural populations subjected to different conditioning temperaturesABSTRACT. The reproductive cycles of Mytilus chilensis vary latitudinally. This species has reduced genetic and morphological differentiation due to its high potential for dispersal. Broodstocks from Yaldad Bay (Chiloé) and Zenteno Bay (Punta Arenas) were conditioned at 9 ± 0.5°C and 15 ± 0.5°C, and were fed a diet (1:1) of Isochrysis galbana and Chaetoceros neogracile. We expected to determine whether conditioning at different temperatures produces changes in the reproductive potential of the populations. Gonadal development was lowest in the broodstocks conditioned at 9ºC, and highest in those conditioned at 15°C, from Chiloé. Fertility was greater in broodstocks from Yaldad than in those from Zenteno. Oocyte diameter was greater in broodstocks from Zenteno, and both populations showed larger diameters at 9ºC. Neither the percentage of fertilized eggs nor the percentage of larvae hatching differed significantly between populations at either conditioning temperature. Therefore, it was not possible to establish differences in the reproductive potential of the populations under the conditions studied herein.

SUMMARY: The present study characterizes the reproductive cycle of Venerupis senegalensis (=V. pullastra) from Ria de Aveiro (Portugal) as well as its nutrient storage and exploitation strategy. The reproductive cycle followed a seasonal cycle that correlated negatively with sea surface temperature, and comprised a ripe stage in winter followed by a spawning period that began in late winter and ended in the early summer. This extended spawning may be an advantageous strategy for the species because it ensures a continuous supply of settlers. Gametogenesis began in late summer/early autumn and intensified with the decrease in temperature during autumn. The condition index increased even during the spawning period, which indicates that there is rapid recovery and that reserves are accumulated during late summer and used later in the gametogenic process. Proteins did not contribute significantly to gametogenesis and the glycogen pattern is typical of conservative species, since gametogenesis depends largely on the amount of glycogen stored. The lipid storage and utilization cycle showed that gametogenesis took place in autumn/winter and that energy reserves were accumulated in summer.Keywords: pullet carpet shell, Venerupis senegalensis, biochemical composition, condition index, reproductive cycle.RESUMEN: Actividad reproductiva y composición bioquímica de la almeja babosa Venerupis senegalensis (Gmelin, 1791) (Mollusca: Bivalvia) de la Ría de Aveiro (costa noroeste de Portugal). -El presente estudio caracteriza el ciclo reproductivo de Venerupis senegalensis (= V. pullastra) en la Ría de Aveiro (Portugal) así como su estrategia de almacenamiento y explotación de los nutrientes. El ciclo reproductivo de esta especie sigue un ciclo estacional que se correlaciona negativamente con la temperatura del agua del mar, que coincide con un estado de madurez en el invierno, seguido de un periodo de desove que se inicia a finales de invierno y termina a principios del verano. Este periodo de puesta prolongado puede ser una estrategia ventajosa para la especie, ya que garantiza un suministro continuo de larvas. La gametogénesis comenzó a finales del verano/inicios de otoño y se intensificó con la disminución de la temperatura durante el otoño. El aumento del índice de condición, todavía durante la puesta, indica una recuperación rápida y una acumulación de reservas a finales del verano, que serán utilizadas en el siguiente proceso de gametogénesis. Las proteínas no contribuyen significativamente a la gametogénesis y el ciclo de glucógeno es típico de las especies conservadoras, una vez que la gametogénesis depende, en gran medida, de la cantidad de glucógeno almacenado. -To develop selection programs for the great scallop, artificial reproduction of the species needs to be improved by gamete cryobanking. Here, a set of four experiments was designed in order to define the basic elements of a cryopreservation protocol for scallop sperm, including extender composition (experiment 1), cryoprotectant selection (2), cooling rate (3), and assessment of the effects of sperm cryopreservation on sperm motility and fertilization capacity (4). Sperm was collected after serotonin injection (100 µl of a 10-mM solution) and frozen in 500-µl straws. For the first three experiments, the percentage of motile fresh sperm (80 ± 4%, mean ± SEM) was significantly higher than that observed for thawed sperm (11 ± 2%). During the first experiment, no significant difference of the percentage of motile thawed sperm was observed among the three saline extenders tested: seawater, calcium-free Hanks' balanced salt solution (Ca-free HBSS) and DCSB4 solution. However, a complementary experiment (2) showed that a significantly higher percentage of motile thawed sperm was recorded using DCSB4 than with Ca-free HBSS as an extender. During the third experiment, sperm motility was higher when using polyethylene glycol (10% PEG) as a cryoprotectant, than when using dimethyl sulphoxide, DMSO (10 and 20%), methanol (10%) and ethylene glycol (10% EG). A higher D-larval rate was obtained during the fourth experiment, using fresh sperm than thawed sperm, and using a 500:1 sperm-to-egg ratio compared with a 50:1 ratio. There is some evidence of inter-individual variations in sperm tolerance to cryopreservation. In conclusion, the highest survival of great scallop sperm after thawing was recorded using the following conditions: DCSB4 extender (1:3 vol/vol sperm to extender dilution), PEG cryoprotectant (10%) and straws maintained at 5.5 cm above liquid nitrogen.
