Reproductive and developmental toxicity studies of toluene. I. Teratogenicity study of inhalation exposure in pregnant rats.

Abstract: Toluene is a widely used solvent in industry which is the subject of abuse among the younger generation. A teratogenicity study of toluene by inhalation exposure was carried out in Sprague-Dawley rats and the effects on dams, fetuses and offspring were assessed. Pregnant females were exposed to 600 or 2000 ppm toluene for 6 h/day from day 7 to day 17 of pregnancy. The control group inhaled conditioned clean air under the same exposure conditions. Maternal exposure to 2000 ppm toluene caused significant toxic e… Show more

“…With these exposure patterns to males or females, there were no significant effects on fertility or reproductive performance, but fetal growth was significantly affected [Roberts et al 2003]. A similar pattern of exposure used by Ono et al [1996] also had no effect on fertility, but these studies do not model patterns of exposure typical of solvent abuse.…”

“…Subacute exposure of male rats to a high level (2000 ppm) of toluene vapor can elicit mild toxic changes in the kidneys, thymus, and reproductive organs of males. The results demonstrated that toxic effects on the fertility and reproduction were not only in females but also in males exposed to toluene vapor 10) . In male rats thinner, particularly the components ethyl acetate and xylene, was reported to interfere with the functions of the testes and accessory reproductive organ; but toluene has no effect on these functions 11) .…”

“…The reproductive and developmental toxicity of toluene in animals and man is well characterized (Donald et al 1991;Ono et al 1996). Studies have shown that in rats toluene just like ethanol, has toxic effects on the brain development as well as disturbances in the cognitive functions (Hass et al 1999).…”
